Insomnia,as one of the diseases with the highest incidence in today's fast-paced life,has attracted more and more attention from the medical community.Through long-term clinical observation,insomnia of liver depression syndrome accounts for a large proportion of insomnia.But how does "relieving qi stagnancy in liver",a long-term treatment for insomnia of liver depression syndrome,play a therapeutic role,and how does it affect the mTOR signaling pathway? At present,the research is not clear.In this study,animal behavioral experiments,Western Blot and Real Time PCR were used to detect the changes of behavior and PI3K/Akt/mTOR signaling pathway related protein expression in hippocampus of insomnia rats with liver depression syndrome,and to provide objective basis for the treatment of insomnia with liver depression syndrome by " relieving qi stagnancy in liver " method,and to explore its specific mechanism.The specific research contents are divided into two parts: theoretical research of Chinese medicine and experimental research.This paper mainly expounds the understanding of the etiology,pathogenesis,diagnosis and treatment of insomnia in Chinese medicine and its relationship with the study of modern cell autophagy.Taking the representative mTOR signaling pathway in modern cell research as the breakthrough point,the therapeutic effect and specific mechanism of Xiaoyao San on insomnia of liver depression syndrome are studied.AIM: To duplicate the insomnia rat model of liver-depression syndrome,observe and compare the behavioral changes and the expression differences of related proteins in hippocampus of rats in each group,and study the effect of Xiaoyao San on the behaviour of insomnia rats with liver-depression syndrome,and the effect of Xiaoyao San on the mTOR signaling pathway of rats in each group.To explore the effect mechanism of relieving qi stagnancy of liver in treating insomnia of liver depression syndrome by changing the expression of related proteins in this signaling pathway.Methods: Healthy male SD rats were purchased and randomly divided into model group and blank group.Rat models of insomnia of liver depression syndrome were established by chronic tail-clamping stimulation and intraperitoneal injection of compound factors of PCPA.The blank group was fed normally.Then the model rats were randomly divided into model group,high-dose group,medium-dose group,low-dose group,Tianwang Buxindan group and Western medicine group.There were seven groups in the blank group.From the 7th day of modeling,the rats in each group were given drugs for 14 consecutive days.The corresponding doses of Xiaoyao San were used to intervene in each Chinese medicine group,Tianwang Buxindan group was intervened with Tianwang Buxindan decoction,Western medicine group was intervened with diazepam;after animal behavior experiment,all rats were anesthetized with10% chloral hydrate,and the hippocampus was removed.Then the expression of mTOR signaling pathway related proteins in hippocampus was detected by WB and Real-Time PCR,and the effect of Xiaoyao San on the signaling pathway was studied.Result: The frequency of facial washing and hair grooming in the model group and Tianwang Buxindan group decreased significantly.After further simulating the insomnia model of liver depression syndrome,the circadian rhythm of the model group basically disappeared.In the animal behavior experiment,The model group and Tianwang Buxindan group,the activity degree and activity frequency of rats were significantly reduced,and the activity degree and activity frequency of rats in each drug treatment group were increased.Compared with the model group,the expression of PI3 K,Akt and mTOR in the hippocampus tissue of rats in each treatment group was up-regulated,and the effect of the middle dose group and the western medicine group was significant.CONCLUSION: According to the observation of macroscopical signs and the data of animal behavior experiment,the insomnia rats with liver depression syndrome showed different degrees of liver depression and insomnia.After drug intervention,the related symptoms of rats in each drug treatment group were alleviated to varying degrees,but the improvement effect of Tianwang Buxindan was not obvious;WB,PCR and other research methods showed that Xiaoyao San can obviously improve the related symptoms of insomnia rats with liver-depression syndrome and has a good therapeutic effect on insomnia rats with liver-depression syndrome.Its specific mechanism may be through activating PI3K/Akt/mTORsignaling pathway to achieve the goal of regulating nervous system,repairing nerve injury and treating insomnia of liver-depression syndrome.
