Objective:To observe the effect of shenlingbaizhu powder modified compound on the expressions of AKT and P70S6 in mice with bone cancer pain,and to determine the efficacy of shenlingbaizhu powder modified compound in the treatment of bone cancer pain and explore its possible mechanism.Methods:Fifty-eight male BALB/c mice were randomly selected,9 from the normal group,and the remaining 49 were used to establish the cancer pain model of lung cancer bone metastasis.During the modeling process,4 died.The remaining 45 model mice were divided into the model group according to the random number table method.Normal group,model group normal feeding;In the high,medium and low dose groups of shenlingbaizhu powder,0.4ml per day was given to each mouse at week 6 after model establishment for 7consecutive days.The rapamycin group was given intraperitoneal dose of4mg/kg once a day for 7 days at week 6 after model establishment.Modeling was evaluated by behavioral observation,HE staining and pain behavioral test.Tunel staining was used to observe tumor cell apoptosis.Immunohistochemistry and Western blot were used to detect the protein expressions of AKT and P70S6 in the tibia of mice in each group.Results:1.According to the behavioral changes of mice in each group,the comparison of HE pathological detection between the normal group and the model group,as well as the comparison of pain behavior of mice before and after modeling suggested that the model was successfully established.2.The results of pain behavior test after administration showed that,comparedwith the model group,the pain threshold of mice in the medium and low dose groups of shenlingbaizhu powder and rapamycin group was significantly increased(P < 0.01).There was no significant difference between rapamycin group and medium and low dose group(P > 0.05).3.HE test after administration showed that the number of tumor cells in the bone marrow cavity of the different dose groups of shenlingbaizhu powder was decreased compared with that of the model group,and the bone marrow cells in the bone marrow cavity of the medium dose group were roughly uniform,with a small number of tumor cells and slight changes in bone structure.In the rapamycin group,the number of tumor cells in the bone marrow cavity was decreased,but there was significant hemorrhage.4.Immunohistochemical results showed that,compared with the model group,the positive rates of AKT and P70S6 in the high,medium and low dose groups and rapamycin groups were all decreased(P < 0.05).Compared with the rapamycin group,the positive rates of AKT and P70S6 proteins in the medium dose group were lower(P < 0.01).5.Tunel results showed that compared with the model group,the apoptosis values of tumor cells in the high and medium dose groups of shenlingbaishu powder and rapamycin group were significantly up-regulated(P < 0.01).There was no significant difference between rapamycin group and medium dose group(P > 0.05).6.Western blot analysis showed that,compared with the model group,the AKT protein expressions of shenlingbaizhu powder in the high,medium and low dose groups and rapamycin groups were all decreased(P < 0.01).The protein expression of P70S6 in medium and low dose group and rapamycin group was lower than that in model group(P < 0.05).Conclusion: 1.The mouse model of cancer pain caused by bone metastasis of lung cancer was successfully established.2.Shenlingbaizhu powder modified compound can alleviate the pain symptoms of bone cancer pain mice and promote the apoptosis of tumor cells.3.Shenlingbaizhu powder modified compound can reduce the expression of AKT and P70S6 protein in the tibial tissue of mice with bone cancer pain.4.Shenlingbaizhu powder modified compound may inhibit the expression of AKT and P70S6,thereby affecting the formation of tumor blood vessels and tumor tissues,and playing a role in the treatment of bone cancer pain.
