Objective:To observe the effects of Chaoyi Taiyin Tiaowei Decoction combined with Xiongdan Powder on oxidative stress,inflammatory response,neurological deficit and brain histomorphology in Middle Cerebral Artery Occlusion(MCAO)Model rats.To study the neuroprotective effects of Chaoyi Taiyin Tiaowei Decoction combined with Xiongdan Powder on the rat model of Middle Cerebral Artery Occlusion(MCAO)Model.To provide experimental support for the clinical treatment of ischemic stroke.Methods:Thirty healthy male SD rats were randomly divided into sham operation group,model control group,low dose Chaoyao group,high Chaoyao dose group,and 5 groups of positive control group with western medicine.Except for the sham operation group,where only the antenor cervical skin was cut and the unilateral common carotid artery was bluntly isolated.The other 4 groups of rats were induced by cerebral ischemia reperfusion in rats by the Middle Cerebral Artery Occlusion(MCAO)Model.Method of administration:(1)Sham operation group:normal saline 1 ml/100 g/d.(2)Model control group:normal saline 1ml/100g/d.(3)High Chaoyao dose group:Taiyin Tiaowei Decoction combined with Xiongdan Powder 13.96g/kg/d.(4)Low dose Chaoyao group:Taiyin Tiaowei Decoction combined with Xiongdan Powder 3.49g/kg.(5)Western medicine positive control group:Nimodipine 0.0125g/kg/d.Continuous administration for 14 days.After administration,the degree of neurological deficit was assessed by Longa neurobehavioral score.Histopathological changes were observed by HE staining after brain tissue section.Serum interleukin-10(IL-10)content,oxide dismutase(SOD),malondialdehyde(MDA),tumor necrosis factor(TNF-a)were determined by enzyme-linked immunosorbent assay.Results:1.The results of neurological impairment evaluated by Zea-Longa score:The scores of the other four groups were improved compared with pre-gavage except the sham operation group,and there was statistical significance(P<0.05).After 14 days of gavage,compared with the model group,the western medicine control group,the high Chaoyao dose group and the low Chaoyao dose group had improved scores(P<0.05).Among them,the western medicine control group and the high Chaoyao dose group unproved significantly.2.HE staining results:it was observed that the damage degree of neurons in the high Chaoyao dose group,the low Chaoyao dose group and he Western medicine group was less than that in the model group,and the improvement was more obvioushe high Chaoyao dose group and the Western medicine group.3.The effects on serum TNF-? and IL-10:After intragastric administration,compared with the model group,the levels of TNF-a in the other three groups were decreased(P<0.05).The levels of IL-10 in serum of Western medicine group,the low Chaoyao dose group and the high Chaoyao dose grouplow Chao prescription group were improved in varying degrees.4.The effects on serum SOD and MDA:Compared with the model group,the serum SOD of the western medicine group,the high Chaoyao dose group and the low Chaoyao dose group were increased(P<0.05).There was no significant difference between the western medicine group and the high Chaoyao dose group(P>0.05).Serum MDA levels were all decreased(P<0.05).The changes in the western medicine group and the high Chaoyao dose group were more significant.There was no significant difference between the western medicine group,the high Chaoyao dose group ana toe low Cnaoyao dose group(P>0.05).Conclusion:Taiyin Tiaowei Decoction combined with Xiongdan Powder can improve neurological dysfunction in rats with cerebral ischemia-reperfusion,and can effectively scavenge free radicals,Increase the activity of SOD,reduce serum MDA levels,decrease TNF-? levels increase IL-10 levels and reduce post-ischemic inflammatory response.
