Objective:To investigate the effects of taurine on blood glucose,blood lipids,oxidative stress and insulin-related signaling pathways in liver and kidney of type 2 diabetic rats,and to investigate the regulation of taurine on glucose and lipid metabolism and oxidative stress in type 2 diabetes mellitus.Research on the mechanism of regulation.Methods:A rat model of type 2 diabetes was established by feeding healthy male SD rats with high-sugar and high-fat diet and low-dose intraperitoneal injection of 35 mg/kg streptozotocin(STZ).Were randomly divided into model group,Metformin(400 mg/kg),taurine high-dose group(600 mg/kg)and low dose taurine(400 mg/kg),in addition to the normal group was fed an ordinary diet,the rest of The rats were given high-sugar and high-fat diets.After 7 weeks of continuous gavage,fasting blood glucose(FBG),triglyceride(TG),total cholesterol(TC),high-density lipoprotein(HDL),and low were measured in each group.Density lipoprotein(LDL)and very low density lipoprotein(VLDL),liver and kidney function,and serum insulin(Ins)were calculated for insulin resistance index(IRI)and insulin sensitivity index(ISI).Determination of superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT)and glutathione(GSH),total antioxidant capacity(T-AOC)in liver and kidney tissues using a kit.Detection of phosphatidylinositol-3-kinase(PI3K),protein kinase(Akt),glucose transporter(GLUT4),Keleh-like epichlorohydrin-related protein-1(Keapl)in liver and kidney tissues by RT-PCR Expression ofheme oxygenase-1(HO-l),quinine oxidoreductase 1(NQO1),and nuclear factor E2 related factor 2(Nrf2)mRNA.Western blotting(Western-Blot)was used to detect PI3K,Akt and GLUT-4 protein expression in liver and kidney tissues.Results:Biochemical parameters:high dose of taurine could reduce FBG and FINs in DM rats(P<0.05)and decrease serum TC,TG and HDL-C(P>0.05).Oxidative stress index:In the liver tissue,the high dose of taurine can increase the activity of SOD,CAT and T-AOC(P<0.05)and decrease the content of MDA(P<0.05).Low-dose taurine group increased T-AOC activity and decreased MDA content(P<0.05).In the kidney tissue,the high dose of taurine increased SOD,GSH,T-AOC(P<0.05)and decreased MDA(P<0.05).mRNA and protein detection results:the high dose of taurine can up-regulate the liver,The levels of PI3K Akt,GluT4 mRNA and protein in renal tissues(P<0.05),the high dose of taurine up-regulated the mRNA levels of Nrf2,HO-1 and NQOl in liver and kidney tissues(P<0.05),and down-regulated the mRNA of Keap1.Level(P<0.05).Conclusion:Taurine can reduce blood glucose,participate in lipid metabolism,alleviate insulin resistance and improve oxidative stress in type 2 DM rats.The mechanism may be related to activation of liver and kidney tissues P13K-AKT-GLUT4 and Keap1-Nrf2-ARE signaling pathway is involved.
