Object: To investigate the effects of renal ischemia-reperfusion injury on myoc-ardium of isolated organs and its mechanism by observing the expression of HMGB1-TLR4-MyD88-NF-kappa B signaling pathway and inflammatory factors IL-6,IL-10 and IL-17 in renal ischemia-reperfusion and myocardial injury of rats.Methods: 16 Healthy male Wistar rats were randomly divided into two groups.The right kidney was removed in both groups.In group C,the left renal artery was not ligated under the left renal artery,and in group IR,the left renal artery was ligated by two lines to establish the model of renal ischemia-reperfusion injury.Blood,kidney and heart were taken after 45 minutes of ischemia and 4 hours of reperfusion.Western Blot,TUNEL,ELISA and real-time PCR were used to detect plasma concentration of Tn-I,HMGB1,IL-6,IL-10 and IL-17.Western Blot was used to detect the expression of HMGB1,TLR4,MyD88,NF-kappa Bp65 and P-NF-kappa Bp65 in cardiomyocytes.Results: Compared with group C,the express-ion of p-NF-kBp65 and TLR4 protein in group I/R was significantly increased(P<0.05).Compared with group C,the levels of IL-17 A and Tn-I in serum of group I/R increased significantly(P<0.05),but the levels of HMGB-1,IL-6 and IL-10 d-id not increase significantly(P>0.05);the expressions of NF-kappa Bp65 and TLR4 in group I/R increased significantly compared with group C(P<0.05),but the ex-pressions of HMGB1 and MyD88 were not significant(P>0.05);the gene tables o-f MyD88,TLR4 and NF-kappa Bp65 in heart tissue of group I/R were not significant(P>0.05).The expression of HMGB1 gene in group C was significantly highe-r than that in group C(P<0.05),but there was no significant difference between the two groups(P>0.05).Conclusion: Renal ischemia-reperfusion injury can damage isolated myocardium in rats,which may be mediated by HMGB1-TLR4-MyD88-NF-kappa B signaling pathway and inflammatory factors.
