Objective: To researched the quality standard of Xuzaogengping Pills and main Pharmacodynamics.Method:(1)TLC was used to identify Radix Rehmanniae and Semen Ziziphi Spinosae.HPLC to determine the content of flavonol glycosides.Chromatographic column:Symmetry shield Rp C18 column(4.6 mm x 150 mm,5 m),mobile phase consisted of acetonitrile-0.6% phosphoric acid(32:68)at the flow rate of mg·m L-1.The detection wavelength was set at 208 nm.The column temperature was(30±5)?,and the sample size was 10?L on Waters Symmetry shield Rp C18 analytical.(2)Study on toxicology of Xuzaogengping Pills.(3)Rats were established the Climacteric hypertension model,and samples were random Ly divided into model group,Xuzaogengping Pills high dose group,Xuzaogengping Pills low dose group and positive group(Valsartan capsule).Set another blank geoup.Assayed the change of blood pressure,Estrogen,Testosterone,SOD,MDA,NO and e NOS of the rats.Observed the morphological changes of rat vascular tissue and the changes of s GC 1,s GC 1,pkg-1 and p-vasp in rat blood vessels.Result:(1)The characteristic spots of Radix Rehmanniae and Semen Ziziphi Spinosae were detected by TLC.The amount of flavonol glycosides in xuzaogengping pills was determined by HPLC at least 1.20mg/g.(2)There was no obvious acute toxicity in the pills.(3)The blood pressure of rats in the high and low dose groups was decreased obviously;Compared with the model group,the contents of serum estrogen and NO in the positive group,the high dose group and the low dose group were all increased(P<0.05);The serum e NOS content in the positive group and the high-dose group of xuanzao gengping pills was increased(P<0.05);The contents of SOD and MDA in serum of the positive group,the high and low dose groups were all decreased(P>0.05);The content of testosterone in kidney tissue of rats in the positive group was decreased,while the content of testosterone in the high-dose and low-dose groups was increased,but the differences were not statistically significant.(4)The morphological changes of rat vascular tissue were obvious.(5)Compared with the model group,the sGC ?1 subunit protein expression was increased in the positive group and the high dose group;The expression levels of s GC ?1 subunit and p-vasp protein in the blood vessels of rats in the positive group,the Xuzaogengping pill group and the low-dose group were increased to varying degrees,and the expression range was widened;The expression of pkg-1 protein in the blood vessels of the positive group,the high dose and the low dose groups decreased to different degrees.Conclusion: The quality standard of Xunzaogengping pills was established.This method is convenient,reliable and can effectively monitor and evaluate the quality of Xunzaogengping pills.Xuanzaogengping pills has a certain therapeutic effect on menopausal hypertension,and its antihypertensive mechanism may be related to NO/s GC/c GMP/PKG.
