Objective: To prepare a targeted ultrasound contrast agent carrying CXCXL12 antibody,establish a nude mouse ovarian cancer SKOV3 xenograft model and perform pathological staging to achieve targeted recognition of CXCL12 in ovarian cancer xenografts in vivo,explore the imaging effect of this contrast agent on different pathological stages of ovarian cancer xenografts in nude mice and the correlation between its contrast parameters and in vitro markers.Methods: A targeted ultrasound contrast agent carrying CXCL12 antibody was prepared,and a subcutaneous xenograft model of 50 nude mouse SKOV3 cells ovarian cancer was established.One group was randomly selected on the 10 th,20th,30 th,40th,and 50 th day after transplantation,and the selected nude mice were subjected to general and targeted contrast-enhanced ultrasound imaging,then nude mice were sacrificed and pathological staging of transplanted tumor tissues was performed.The expression of CXCL12 RNA in tumor tissues was detected by RT-PCR.The correlation between the expression of CXCL12 and peak intensity(PI)and time to peak(TTP)of target contrast parameters in transplanted tumors of different pathological stages was analyzed.Results: According to the results of blind reading,13,13 and 11 specimens with pathological stages I,II and III were obtained.The PI value of targeted contrast-enhanced ultrasound in stage I and II ovarian cancer transplanted specimens was higher than that of conventional contrast-enhanced ultrasound.The TTP value of targeted contrast-enhanced ultrasound in stage I ovarian cancer transplanted specimens was lower than that of conventional contrast-enhanced ultrasound,the difference was significant(P < 0.05).There were differences in the expression of CXCL12 RNA between differentpathological stages of ovarian cancer xenografts,Statistical significance(P<0.05),and stage III expression was the highest.The relative expression of CXCL12 RNA in stage II and III ovarian cancer xenografts was positively correlated with PI(P<0.05).Stage I and III ovarian cancer xenografts CXCL12 The relative expression of RNA was positively correlated with the targeted contrast parameter TTP(P<0.05).Conclusion: Targeted contrast-enhanced ultrasound with CXCL12 can provide new ideas and targets for early diagnosis of experimental ovarian cancer.
