Objective: To observe the expression of matrix metalloproteinase-3 and matrix metalloproteinase-13 in cartilage and synovium of patients with knee osteoarthritis of different TCM syndromes,and explore the correlation between the above indicators and TCM syndromes of knee osteoarthritis,and provide molecular biological evidence for the connotation of TCM syndromes.Methods: Fifteen patients with KOA and joint replacement were selected from the Affiliated Hospital of Traditional Chinese Medicine of Xinjiang Medical University from January 2018 to December 2018,at the same time,5 healthy and non-osteoarthritis patients who underwent fracture repair,amputation and arthroscopic surgery after trauma were selected,and then collect the synovium and cartilage during the operation.The expression of synovium and cartilage was detected by RT-PCR and Western-Blot.Results: There was no significant difference in the expression of MMP-13 in synovium and cartilage between KOA patients and control group.(P > 0.05,no significant difference).Detection of MMP-3 in synovium of KOA patients showed that there were significant differences between kidney deficiency and blood stasis type and blood stasis blockade type and control group.(P < 0.05,the difference was significant).MMP-3 was highly expressed in wind-cold obstruction type,blood stasis obstruction type and kidney deficiency and blood stasis type.MMP-3 in cartilage tissue of KOA patients was detected that there was significant difference between the blood stasis blocking type and kidney deficiency and blood stasis type and the control group,and MMP-3 expression was low in the three TCM syndromes groups.Conclusion: MMP-3 is specifically expressed in cartilage and synovium of KOA patients and plays a role in the development of KOA.The expression of MMP-3 in cartilage of KOA patients with kidney deficiency and blood stasis is lower than that of the other two types,which can provide objective basis for TCM syndrome differentiation and typing of knee osteoarthritis.
