With the development of urban process,the acceleration of life rhythm and the increase of job competition pressure,insomnia has become a major problem faced by most urban population.At present,the main clinical treatment of insomnia is drug treatment and non-drug treatment.Drug therapy can promote sleep quickly,but it has certain side effects and drug dependence.Long-term use of this drug is harmful to human health.Among the non-drug treatment methods,the traditional Chinese acupuncture treatment for insomnia,especially ear acupuncture,has shown good clinical efficacy by needling the vagus nerve of the concha auris.In addition,the acupuncture treatment also has many advantages,such as no side effects,no dependence and convenience.However,the mechanism of acupuncture for insomnia has not been fully elucidated,especially in the study of EEG rhythm changes.The clinical analysis of EEG signals mostly stays in time domain,lacking the EEG characteristics of power spectrum and non-linear dynamics.Therefore,based on the treatment of insomnia by ear acupuncture,animal experiments and clinical human experiments have been carried out successively in this paper.Firstly,the effect of treatment course on EEG rhythm was explored through animal experiments.The aim is to validate the effectiveness and establish the basis treatment course for clinical trials.Then,based on different EEG sleep stages,the changes of EEG rhythm in insomnia patients before and after treatment and the differences with healthy groups were further studied through clinical human experiments.In the experiment,EEG signals were collected for preprocessing and sleep stages,and then the rhythmic changes of EEG signals were studied from the perspective of power spectrum analysis and non-linear dynamic characteristics.The main results of this paper are as follows:(1)Stimulation of auricular concha vagus nerve by Acupuncture can increase the slow wave component and decrease the fast wave component in the EEG signal of rats,thus reducing the frequency of the center of gravity of EEG and improving sleep quality.After 6-7 days of continuous acupuncture,there is a significant difference between the treatment group and the insomnia group(p<0.05),and the power spectrum distribution is basically stable after four days of continuous acupuncture treatment.(2)Entropy and complexity of EEG signal in insomnia group were much higher than those in normal group.With the increase of treatment days,the values of entropy and complexity in treatment group decreased gradually,approaching the normal level,among these groups,the differences of sample entropy,LZ complexity and CO complexity were the greatest(p<0.05).(3)Most insomniacs have fewer nighttime awakenings and higher sleep efficiency after acupuncture treatment,which indicates that acupuncture treatment has a certain effect on improving sleep quality.(4)After treatment,the power spectrum distribution and non-linear dynamic characteristics of insomnia patients are closer to the level of healthy control group than before treatment,and the difference is generally reduced.Acupuncture therapy can effectively regulate EEG rhythm,improve EEG complexity and improve sleep quality.
