?Objective? The explore the different surface defect of subchondral bone on the early tissue morphology of articular cartilage.?Methods? Opened bone window at entocondyle of right tibia which is about 3mm below tibia platform;applied excochleation of subchondral bone after drilling corresponding depth in coronal with annular bit.The drilling depths of coronal in group A,B and C respectively were 10,15 and 20mm;meanwhile,the coloboma superficial areas respectively occupied 50%,75% and nearly 100% of total superficial areas of rabbit knee cartilage.The right knee specimen was taken out 6 weeks after operation when killing the animal,so as to observe the overall morphology of right tibia.The pathological section was made with HE dyeing to observe histology morphologic change.The cartilage degeneration pathological Mankin grade was adopted and Pearson was used to analyze the correlation of Mankin grades among the groups.?Results?The cartilage surface in group A was close to normal color in pink,which was smooth;the cartilage surface in group B was rough in drab yellow,and the synovium hyperplasia was obvious;the cartilage surface in group C was rough without luster,the synovium hyperplasia was obvious with significant fracture,and partial cartilage surface collapsed;the Mankin grades in group B and C [(6.30±1.89)?(7.08±2.10)] were all higher than that in group A [(2.70±2.00)](P<0.05);the comparative difference of Mankin grades in group B and C was not statistically significant(P>0.05).Pearson correlation analysis result showed that,the Mankin grade in group A presented positive correlation with that in group B and C.?Conclusion? When the defect surface of subchondral bone was less than or equal to the 50% of total superficial areas,the cartilage only triggered slight degeneration;when the defect surface of subchondral bone exceeded 50% of total superficial areas,the cartilage would generate serious degeneration in earlier stage.
