Objective:To compare the expression levels of gene Micro302A,Micro498 and Micro520E in normal rabbit articular cartilage cells and injured cells.Methods:Rabbit articular chondrocytes were obtained from three weeks ages New Zeland rabbits.Some scratch-wound modes in primary cultured cells were established in vitro.The expression levels of the three particular MicroRNAs in normal cells,and after 1,3,5,and 7 days of injured cells were detected by qRT-PCR technology.Results:Rabbit articular cartilages were successfully separated,and some scratch-wound models in cultured cells were established.Compared with normal group,the results of qRT-PCR showed that the expression level of Micro302A increased obviously on the 1st day after injury,reached the peak on 3rdd days,and kept the decreasing until the 7th days.There was no obvious change of Micro498 expression level on the 1st day,but rised,and touched the highest point on 3rdd day with sharp increasing before decreasing to the same level of normal cells on 7th days.The expression level of Micro 520E increased on first day,then began to decrease on 3rd,which was lower than that in normal cells on the 7th day.Conclusions:There is statistical significance in the expression levels of Micro 302A,Micro 498 and Micro 520E between normal rabbit articular cartilage cells and injured cells after 1 to 7 days,which refered to different functions in the healing processes of articular cartilage cells after injured.The results provide important the experimental basis for using MicroRNA to repair the injured articular cartilage in the future.
