Objective:To investigate the expression of Cancerous Inhibitor Of Protein Phosphatase-2A(CIP2A),matrix metallopr-oteinase-2(MMP-2)and CD44v6 in papillary thyroid carcinoma(PTC),thyroid adenoma,nodular goiter and normal tissue adjacent to papillary thyroid carcinoma,and to explore the expression of CIP2A,MMP-2 and CD44v6 in different thyroid tissues,whether there have differences of expression,and whether the expression is related to the clinical pathological parameters of papillary thyroid carcinoma,the results of this study may provide new experimental basis for clinical treatment of papillary thyroid carcinoma.Methods:1.Source of data:The paraffin tissue samples from patients with thyroid disease that were surgically treated in the Department of Oncology at the Second Affiliated Hospital of Jiaxing University from January 2014 to November 2016 were collected.All enrolled patients met the criteria of inclusion and exclusion for this study.2.Detection index:The expression of CIP2A,MMP-2 and CD44v6 in 67 cases of papillary thyroid carcinoma,26 cases of thyroid adenoma,23 cases of nodular goiter and 22 cases of normal tissue adjacent to papillary thyroid carcinoma were detected by immunohistochemical method.The sex,age,gland outside infiltration,lymph node metastasis,tumor diameter size,and pTNM staging were recorded in all enrolled patients.3.Data analysis:For the analysis of the expression levels of CIP2A,MMP-2,and CD44v6 proteins,the product of two scoring methods was used as the final result;the relationship between the expression of CIP2A,MMP-2,and CD44v6 and the clinicopathological parameters in patients with papillary thyroid carcinoma were analyzed.The counting data was analyzed with?~2 test;the correlation analysis of CIP2A,MMP-2 and CD44v6 were performed using SPSS 19.0 software,and calculation the correlation coefficient.The difference was considered statistically significant at P<0.05.Results:1.A total of 138 patients were included in this study.Among them,67 cases of papillary thyroid carcinoma,26 cases of thyroid adenoma,23 cases of nodular goiter,22cases of normal tissue adjacent to papillary thyroid carcinoma.Among 67 cases with papillary thyroid carcinoma patients,male was 14 cases and females was 53 cases,the age ranged from 23 to 82 years old and the median age was 50 years old.2.Immunohistochemical staining showed that the positive expression of CIP2A and MMP-2 was mainly located in the cytoplasm,CD44v6 was mainly expressed in the cell membrane,and a small amount was expressed in the cytoplasm.And the expression of their in papillary thyroid carcinoma was significantly higher than that in thyroid adenoma,nodular goiter and normal tissue adjacent to papillary thyroid carcinoma.3.The product of the two methods as the final results showed that the positive expression rate of CIP2A in papillary thyroid carcinoma,thyroid adenoma,nodular goiter and normal tissue adjacent to papillary thyroid carcinoma was 71.64%,30.77%,21.74%and13.64%respectively,P<0.05.The positive rate of MMP-2 expression was 80.60%,46.15%,30.43%and 18.18%respectively,P<0.05;the positive expression rate of CD44v6 was 76.12%,23.08%,21.74%,and 13.64%respectively,P<0.05.4.Apply?~2tests to analyze the relationship between CIP2A,MMP-2,and CD44v6 expression levels and clinicopathological parameters such as sex,age,tumor diameter size,gland outside infiltration,lymph node metastasis,and pTNM staging in patients with papillary thyroid carcinoma.The expression of CIP2A and CD44v6 was only significantly associated with lymph node metastasis(P<0.05),The expression of MMP-2 was only significantly associated with pTNM staging(P<0.05).5.Correlation analysis was used between CIP2A,MMP-2 and CD44v6 showed that the correlation coefficient r between CIP2A,MMP-2 and CD44v6 was(0.11),(P>0.05);(0.06),(P>0.05);the correlation coefficient r of CIP2A and MMP-2 expression levels was(0.02),(P>0.05).Conclusion:1.Overexpression of CIP2A,MMP-2 and CD44v6 in papillary thyroid carcinoma may be a clinical marker for the combined diagnosis of papillary thyroid carcinoma.2.The expression of CIP2A is associated with the lymph node metastasis in papillary thyroid carcinoma,It is suggesting that overexpression of CIP2A may promote lymph node metastasis of papillary thyroid carcinoma.The high expression of MMP-2is associated with the pTNM staging of papillary thyroid carcinoma.And there was also exist correlation between the expression of CD44v6 and lymph node metastasis of papillary thyroid carcinoma.CIP2A,CD44v6 and MMP-2 may provide a new direction of therapeutic strategies for papillary thyroid carcinoma.3.There was no correlation between the expression of CIP2A,MMP-2 and CD44v6 in thyroid papillary carcinoma,it is suggesting that there may be no synergistic relationship between CIP2A,MMP-2and CD44v6 in the development of papillary thyroid carcinoma.
