Objective:To discuss the correlation of Galectin-3,IL-6,MCP-1 and human atherosclerotic plaque stability.Methods:Sixty patients visited the vascular surgery of Affliated Yantai Yuhuangding hospital of Qingdao University in March 2016 to December 2016 were recruited in this study.Among them,38 cases were diagnosed as carotid atery stenosis by carotid artery color ultrasonography examination.These 38 cases were divided into stable plaque group(n=21)and unstable plaque group(n=17)based on standard of AHA(American Heart Association);22cases without carotid atery stenosis based on carotid artery color ultrasonography examination were regarded as control group.Clinical data of all patients were recorded in detail.Venous blood of patients was collected from elbow vein and serum was separated.Detection of the level of Galectin-3,IL-6,MCP-1 in serum were carried out by double antibody sandwich enzyme-linked immunosorbent(ELISA).The rest of the indexes were detected by the hospital clinical laboratory.Femoral artery intima tissue of patients with carotid artery stenosis were obtained during carotid intima peeling technique(carotid endarterctomy).Femoral artery intima tissue of patients without carotid artery stenosis were obtained femoral artery dissection(patients received with other reasons).Quantitative and locative detection detection the expression level of Galectin-3,IL-6,MCP-1 of femoral artery intima tissue of patients with or without carotid artery stenosis by immunohistochemical.And reassured the expression level of Galectin-3,IL-6,MCP-1 of femoral artery intima tissue of patients with or without carotid artery stenosis by Western Blot.Results:Age,sex.body mass index,smoking history,history of hypertension,diabetes,indicators have no statistical difference between three groups(P>0.05)through statistical analysis of the general clinical data.The level of high density lipoprotein(HDL-ch)of normal control group was higher than that of the stable plaque group and the unstable plaque group(P<0.05)based on blood routine and biochemical indexes of blood examination.There were no significant differences in the rest of the indicators.By ELISA method,the level of Galectin-3 in the normal control group,stable plaque group and unstable plaque group were 452.5± 141.49(pg/ml),938.88±425.07(pg/ml).1886.54±453.88(pg/ml)respectively(P<0.05).The level of IL-6 inin the normal control group,stable plaque group and unstable plaque group were 63.95±21.01(ng/L),123.31 ±53.86(ng/L),123.31 ±53.86(ng/L)respectively(P<0.05).The level of MCP-1 in the normal control group,stable plaque group and unstable plaque group were 204.04±65.16(ng/L),368.9± 135.92(ng/L),711.6± 130.88(ng/L)respectively(P<0.05).All in all,the level of Galectin-3,IL-6.MCP-1 in stable plaque group and unstable plaque group were obviously higher than that in normal contol group(P<0.05).By Western Blot analysis,the expression level of Galectin-3,IL-6 and MCP-lin stable plaque group and unstable plaque group were higher than that in normal control group.And the expression level of Galectin-3,IL-6 and MCP-1 in the unstable plaque group level was the highest(P<0.05).By immunohistochemical methods,the level of Galectin-3,IL-6 and MCP-1 were obviously different in the three groups.They were the lowest in the normal control group.Deepest dyeing occured in unstable plaques group,means expression level was highest.The positive rate of Galectin-3 were18.18%(4/22),57.14%(12/21)and 70.59%(12/17)respectively in the normal control group,stable plaque group and unstable plaque group.The positive rate of IL-6 arwere 22.73%(5/22)?61.90%(13/21)?70.59%(12/17)respectively in the three groups.The positive rate of MCP-1 were 22.73%(5/22)?61.90%(13/21)?76.47%(13/17)respectively.Difference between three groups has statistical significance(P<0.001).Conclusions:The level of Galectin-3,IL-6 and MCP-lin serum and of atherosclerosis patients are raised,especially in patients with unstable plaques.Therefore,Galectin-3,IL-6 and MCP-1 all may be involved in the process of the occurrence and development of atherosclerosis.Galectin-3,IL-6 and MCP-1 are closely related to the stability of atherosclerotic plaquyes.They are expected to become an important index of clinical prediction of acute cardiovascular events.However,there are further study to do.
