Objective: This study applied the transcriptome sequencing technology to explore gastric neuroendocrine carcinoma(GNEC)transcriptome landscape,and focus on Neuro D1,the specificity increasing expressed gene of GNEC,to explore its expression and its function in the biology process of GNEC.Method: This study applied the new generation of high-throughput sequencing technology RNA-seq to detect the transcriptome of tissue samples;the RT-q CR and western blot were used to detect the m RNA and protein expression of Neuro D1;using immunohistochemical method to detect Neuo D1 expression in 50 GNEC patients’ paraffin tissue section and conduct relevant prognostic analysis;using CCK8 and SRB method to detect cell proliferation ability;the tablet clone forming experimental wes uesd to detect cell clone forming ability.Result: This study examined the transcriptome study information of 3 carcinoma tissue3 of GNEC patient and the matched adjacent tissues to carcinoma and 3 cases of gastric adenocarcinoma(GAC)patient,which were matched with the clinical basic situation(such as gender,age,tumor stage)to above ones,and showed the transcriptome landscape;here we found 1248 GNEC specific up-expressed genes and 6 GNEC specific dwn-expressed genes;the GO and some other analysis was performed for these GNEC specific expressed genes.One differentially expressed gene,named Neuro D1,had showed that it participated in the multiple processes of nerve and neuroendocrine;firstly we found the m RNA and protein expression were high in GNEC,and immunohistochemical detection of 50 GNEC cases also found that Neuro D1 was able to influence the outcome of patients with GNEC;then we carried on the exploration the function of Neuro D1 in GNEC,and found knock-down of Neuro D1 in GNEC cell line would promote cell proliferation and clone formation while increase the drug sensitivity for 5-fluorouracil.Conclusion: GNEC transcriptome study landscape existed tissue specificity,and its specificity expressed gene Neuro D1 could influence not only the long-term outcome of GNEC patients but also proliferation,clone formation ability and drug sensitivity of GNEC cell. |
