Objective:To investigate the change of nerve growth factor?NGF?in liver and thoracic?T8?spinal dorsal horn?SDH?in rat model of visceral hepatocellular carcinoma pain,and to investigate the neuropathology mechanism of visceral hepatocellular carcinoma pain.Methods:The rat model of visceral hepatocellular carcinoma pain was established by intraperitoneally injected diethylnitrosamine?DEN?.40 male Wistar rats?180±10g?were randomized into experimental group?Group T??n=20?and control group?Group C??n=20?.The rats were injected intraperitoneally with 50mg/kg DEN in Group T and1ml/kg 0.9%NaCl?same volume?in Group C.Injection plan was twice a week at 1stto4thweek,once a week from 5thto 14thweek,and the modeling ended at 20thweek.All rats were given quantitative feed?20g/day/rat?and unlimited drinking during the modeling.The rat visceral pain was determined by hunching evaluation method.2 rats in each groups were selected randomly and executed at the 12th,14thand 16th week,and liver was examined by pathology with HE stain.The left rats were executed at 20thweek,and the liver and T8 spinal were taken out.The content of NGF in liver and T8 SDH were measured by the immunohistochemical staining technique,the result was analyzed by IOD with Image Pro Plus software and the data analyzed by the SPSS 22.0 software.Results:The rat model of visceral hepatocellular carcinoma pain was established successfully.The content of NGF in liver and T8 SDH in group T is higher than Group C.The IOD of liver in group C was 9.36±1.06 and it in Group T was 16.1±1.45.The IOD of T8SDH in Group C was 1.22±0.35 and it in Group T was 4.20±1.76.The differences were statistically significant?P<0.05?.The high expression of NGF in liver mainly was localized in cell cytoplasm of hepatoma carcinoma cell,and it expressed in T8SDH extensively.Conclusion:The content of NGF in liver and T8 SDH of rat model of visceral hepatocellular carcinoma pain are increased.
