Objective:By observing the lipopolysaccharide(1ipopolysaccharide,LPS)induced and vitamin D and glucocorticoid(budesonide)after the intervention,nasal mucosa epithelial cells in vitro culture GRa glucocorticoid receptor m RNA,GRBS m RNA and nuclear factor-KB(nuclear factor kappa B,NF-KB)expression level of change,effect on local regulating cell factors,to explore vitamin D of glucocortico id resistance type chronic nasal sinusitis-whether to have certain treatment function,and treatment with glucocorticoid whether there is a certain role.Methods:Original training in serum-free curing line of nasal mucosa epithelial cell isolation and culture,extend the three or four generations,induced by LPS and add vitamin D,before and after budesonide,using real-time fluorescent quantitative PCR detection GRa m RNA,GRBS m RNA expression level,further using Western blot method to detect the contents of protein expression of NF-KB.Results:LPS(1mg/L)induced the expression of GRb m RNA and NF-KB in the nasal mucosal epithelial cells increased,and the GRb m RNA and NF-KB was decreased after the blocking of budesonide,the expression of GRb m RNA decreased,and the GRa/GRb m RNA ratio increased,and the difference was statistically significant.After the addition of vitamin D+ budesonide,the expression of GRb m RNA and NF-KB decreased further,and the GRa/GRb m RNA ratio was higher,and the difference was statistically significant.Conclusion:This study suggests that vitamin D may reduce the glucocorticoid resistance type chronic nasal sinusitis patients GRBS m RNA and the expression of NF-KB,improve GRa/GRBS m RNA ratio imbalance,which suggests that vitamin D to glucocorticoids resistance,the treatment of chronic nasal sinusitis-have a role,and in conjunction with glucocorticoids effect is better.
