A Study On The Identification Of National Herb In Xinjiang Called Tianping Grass And Analysis Its Fingerprint And Explore Its Anti-breast Cancer Activity

In Xinjiang,there is a national herb that local people called the Tianping grass.The Tianping grass is not only rich in nutrients,but also has a high medicinal value.It can improve the human immune system,treat a variety of human inflammation,prevent and treat a variety of bacteria or viruses caused by infectious diseases.It also has a better effect to maintain normal human physiological activities.But the specific mechanism has not been studied thoroughly.In this study,we first analyzed the ITS sequence of Tianping grass and it was identified as mulgedium tataricum.Mulgedium tataricum is a plant of the genus Mulgedium,it is also known as Lactuca tatarica.It is perennial herbs,and folk mainly eat it as wild food.It has effect on anti-bacterial,anti-inflammatory and it also can relieve pain.Research about mulgedium tataricum has mainly focused on the extraction and antioxidation of total flavonoids and polyphenols from mulgedium tataricum.In this research,mulgedium tataricum was extracted by ethyl alcohol and water.After extracted,it was concentrated by rotary evaporator.And we established the HPLC fingerprint of mulgedium tataricum to analyze the stability of internal components of mulgedium tataricum.We found that internal components of mulgedium tataricum was highly stable.Research had shown that mulgedium tataricum can inhibit the formation of human leukemia cells(HL-60)and human hepatoma cells(SMMC-7721)cultured in vitro and incidence of breast cancer in our country was getting higher and higher.Therefore,we studied the inhibitory effect of mulgedium tataricum on breast cancer cells(MCF-7).The MTT was used to determine the inhibitory effect of mulgedium tataricum alcohol extracted on breast cancer cells,and we found that it had a significant inhibitory effect on MCF-7 cells.In order to explored the mechanism of the inhibition of the MCF-7 by mulgedium tataricum alcohol extracted,we used Western Blot and RT-PCR to detect the expression of IKK? and we found that the expression of IKK? was notably declined after effected by mulgedium tataricum alcohol extracted.It was proved that mulgedium tataricum alcohol extracted could inhibit MCF-7 cells by inhibiting the expression of IKK?.It also proved that mulgedium tataricum could be used as a potential drug to prevent and treat breast cancer.
