Objective:To analyze the correlation between exosomal miR-766-3p/miR-4787-3p and lymph node metastasis of esophageal squamous cell carcinoma,To investigate the proliferation of esophageal squamous cell carcinoma cells miR-766-3p and miR-4787-3p through cell function experiments,The impact of miR-766-3p/miR-4787-3p on prognosis of patients with esophageal squamous cell carcinoma was evaluated.The value of these candidate microRNAs in predicting lymph node metastasis and judging the prognosis of patients with esophageal squamous cell carcinoma was evaluated.Method:1.To collect plasma samples of patients with esophageal squamous cell carcinoma from Fujian Provincial Cancer Hospital,and to detect the expression levels of candidate microRNAs in plasma exosomes and their cell lines using real-time fluorescent quantitative PCR;analyze the tissues using the Mann-Whitney U test;The expression of exosomal candidate microRNAs was correlated with the clinical characteristics of patients with esophageal squamous cell carcinoma(lymph node metastasis,number of lymph node metastasis,TNM stage,infiltration degree,etc.);Logistic Regression was used to construct a predictive model predicting postoperative recurrence and metastasis.Model 1 Contains demographic characteristics and clinicopathological features.Model 2 contains the variables in model 1 and candidate microRNAs.The area under the ROC curves of the two models was compared using Delong et al.2.By constructing human esophageal squamous cell carcinoma cell lines transfected with the above candidate microRNAs,cell transfection efficiency was observed by fluorescence labeling,cell proliferation was detected by CCK-8 assay,cell scratches,and Transwell chamber migration assay was used to detect cell migration and invasion.ability.3.Survival curves of patients with esophageal squamous cell carcinoma were plotted using the Kaplan-Meier method,Log-rank method was used to compare survival rates,and COX regression models were used to analyze tissue and plasma exosomes miR-766-3p and miR-4787-3p in patients with esophageal squamous cell carcinoma.The impact of prognosis.Result:1.The miR-766-3p(P=0.007)and miR-4787-3p expression levels(P=0.003)in patients with lymph node metastasis were significantly higher than those without lymph node metastasis.The area under the curve of Predictive Model 1 was 0.813(95%CI:0.695-0.931);the area under the curve of Model 2 was 0.889(95%CI:0.776-1.00);candidate microRNAs increased the area under the curve that predicts postoperative metastasis(P=0.037).2.CCK-8 found overexpression of miR-766-3p/miR-4787-3p All of them can enhance the cell proliferation ability.With the increase of the expression level,the cell proliferation ability is enhanced(P<0.05).The cell scratches and transwell chamber migration and invasion experiments show that overexpression of miR-766-3p/miR-4787-3p in the control group,cell migration and invasiveness were significantly increased,and the difference in migration and invasive abilities of the different expression levels groups was statistically significant(P<0.05).3.After adjustment for demographic and clinicopathologic features,exosomes(HR=3.197,95%CI:1.033-9.893,P=0.044),and tissue(HR=3.126,95%CI:1.182-8.266,P=0.022)miR High expression of-766-3p is a high risk factor for poor prognosis of patients with esophageal squamous cell carcinoma.Conclusion:Overexpression of miR-766-3p and miR-4787-3p enhances the proliferation,migration and invasion of esophageal squamous carcinoma cells;plasma exosome high expression of miR-766-3p and miR-4787-3p can induce lymph nodes in patients with esophageal squamous cell carcinoma Metastasis,including the above microRNAs model can improve the area under the curve to predict the metastasis of esophageal squamous cell carcinoma;High expression of miR-766-3p in exosomes and tissues is a high risk factor for poor prognosis of patients.
