| Objective:study the correlation between photosensitizer dosage and erythrocyte hemolysis rate and morphology,so as to provide safety basis for photodynamic therapy of magnesium bromide phthalocyanine photosensitizer.Methods:The photosensitizer magnesium bromide phthalocyanine was added into 2%erythrocyte suspension.After red light irradiation at a single wavelength of 660 nm,the hemolysis rate of erythrocytes was measured 24 hours after incubation.The experimental group(MgPcBr-PCT group,magnesium bromide phthalocyanine photosensitizer incubated after red light),photosensitizer control group(magnesium bromide phthalocyanine P group,adding photosensitizer after incubation),light control group(Group L,light only,no photosensitizer),DMSO photosensitizer group and blank control group(group C,without photosensitizer and light).Photosensitizer dose range(1.0,2.5,5.0,and 10)μmol/L,luminous flux density of 637mw/cm~2.Results:The luminous flux density was 637mw/cm~2,the photosensitizer concentration(1.0,2.5,5.0 and 10)μmol/L,the hematopoietic cells in the PCT group,the P group and the DMSO group were significantly hemolytic.The hemolysis rate of the three groups was(2.12±0.56)%,(3.28±2.32)%and(1.82±0.47)%,respectively(23.33±0.80)%,(19.67±0.74)%,(13.16±0.99)%,significantly higher than the same dose And group C(P<0.05).L group and C group compared with red blood cells hemolysis was not obvious(P>0.05).The erythrocyte slides made by PCT group 1.0 and 10.0μmol/L were observed under 100 times microscope.The damage degree of erythrocyte deformation was more than 1.0μmol/L at 10.0μmol/L.Conclusion:Magnesium halide phthalocyanine photosensitizer has erythrocyte toxicity under photodynamic therapy.After 24 hours of incubation,a certain luminous flux density(637 mW/cm2)and a light time(20 min)And the hemolysis rate increased with the increase of the dose of the photosensitizer.When the concentration was<1.0μmol/L,the hemolysis rate was less than 5%,that is,the international standard organization reference evaluation standard. |
