| Objective:To observe the effect of estrogen on trigeminal and serum calcitonin gene-related peptide(CGRP)and nitric oxide(NO)in rats induced by nitroglycerin.Methods:80 clean and healthy female SD rats as the experimental object,randomly divided into experimental group and the blank control group,experimental group subcutaneous injection made the NTG migraine model,subcutaneous injection amount of saline blank control group,the experimental group were randomly divided into again go ovarian group,ovarian + estrogen replacement group and control group.Among them,the rats that went to the ovary + estrogen replacement group were the exogenous estrogens using the estradiol valerate,and the rats in the other groups were fed for 1 week.Experimental subcutaneous injection made the NTG migraine model,the same amount of saline hypodermic blank control group,building successful blood after 4 hours and trigeminal ganglion specimen,using rt-pcr to detect the expression of CGRP in rat trigeminal ganglion cell,using enzyme-linked immunoassay(ELISA)quantitatively detect the content of CGRP and NO.Results:1.To ovarian group rats serum and CGRP content in the trigeminal ganglion and express the highest(373.43±16.66,35.67±0.97),the control rats higher(328.67±18.78,33.40±0.25),to the ovary + estrogen replacement group is lower(177.34±11.89,25.29±0.76).There were statistically significant differences between the groups(P <0.05).2.Between experimental group and ovary rat + estrogen replacement group hasthe highest content of NO in serum(65.38±2.01)and control group of rats content(60.16±4.83)times,to set the minimum(45.52±5.24),ovary comparative differences between groups there is statistical significance(P < 0.05).Conclusion:1.The expression and content of CGRP in the trigeminal ganglion and serum of the blank control group were lower than that in the experimental group,which confirmed the involvement of CGRP in the occurrence of migraine.2.Between the experimental group,compared to ovarian and give pentanoic acid estradiol alternative trigeminal ganglion of rats and the lowest,the expression of CGRP and content in serum estrogen may by reducing the trigeminal ganglion and the expression of CGRP and content in serum and improve the experimental rats migraine symptoms.3.Compared with the experimental group,the number of NO in the ovary +estrogen replacement group was the highest,suggesting that estrogen may increase the occurrence of migraine by increasing the content of NO. |
