Objective:Zoledronic acid(ZOL)has a certain synergistic effect on antitumor drugs such as cyclophosphamide.We designed the experimental treatment with ZOL and anthracycline drug doxorubicin(ADM)to treat MDA-MB-231 cells(ER-,PR-,HER-2-)to investigate whether ZOL combined with ADM treatment can achieve synergistic or additive effects,whether it can increase the inhibitory effect of ADM alone on tumor cell proliferation and induction of apoptosis,and understand whether the combined effect can not increase ADM effect.Methods:ZOL single drug group,ADM single drug group,ZOL combined ADM group,and control group were set in the experiment,and drug experiments were performed on MDA-MB-231 cells.CCK-8 kit was used to detect the inhibition rate of cell proliferation in each drug group;flow cytometry be used to detect the apoptosis of each group.Results:1.The inhibitory rate of cell proliferation are(21.5±1.2)% after treatment with ZOL concentration of 10 ?mol/L,and the inhibition rate are increased with concentration(p<0.05);the effect was(32.1±1.3)% after 48 hours,the inhibitory rate increased with increasing concentration(p<0.05);at a concentration of ZOL 10 ?mol/L,the apoptotic rate was(13.4±1.3)% at 24 hours,with the increase in the concentration of cells The rate of apoptosis was also significantly increased(p<0.05).2.The inhibition rates of MDA-MB-231 cellsa are treated with ADM at concentrations of 1.25,2.5,5and 10 ?mol/L after 24 hours were(14.4±0.8)%,(28.3±1.5)%,(38.1.±2.3)%,(49.3±3.2)%,(p<0.05);The inhibition rate after 48 hours of treatment was(29.1±2.1)%,(49.3±2.7)%,(67.5±3.3)%,(74.6±3.5)%,(p<0.05);after 24 hours of treatment with 1.25 ?mol/L ADM,the apoptotic rate was(15.1±1.2%).Under the same treatment time,the apoptotic rate increased significantly with increasing concentration(p<0.05).3.The inhibition rate of ZOL and ADM combined group was significantly higher than that of ZOL single drug group and ADM single drug group(p<0.05);the apoptosis rate are(25.5±1.7)% at 24 hours after ZOL combined with ADM treatment,while ZOL single drug group It was(13.4±1.3)% in the ADM monotherapy group(15.1±1.2)%,(p<0.05).Conclusion:Both ZOL and ADM could inhibit the growth and induce apoptosis of MDA-MB-231 cells in a concentration-and time-dependent manner.The combination of ZOL and ADM could inhibit the proliferation and induce apoptosis of MDA-MB-231 cells.And the two drugs have an additive effect.The effect of ZOL can increase the effect of ADM on inhibiting cell proliferation and inducing apoptosis.
