Studies On The Metabolism Of Amino Acids For Cultivation Of 293 Cell

293 cell was used to produce biological pharmaceutical(e.g.recombinant proteins),and these pharmaceutical quality will be affected by 293 cell's quality,so we efforts should be on the techniques for cultivating 293 cell on medium.Based on the 293 cell cultures of amino acids,glucose as the research object,by studying the culture medium composition concentration changes,the metabolism of amino acids,glucose metabolism,cell growth curve to do research,and analyze nutrition metabolism of 293 cells.The main research be as follows:(1)6-aminoquinolyl-N-hydroxysuccinimidyl carbamate(AQC)was used as pre-column derivatization reagent,the amino acid content of 293 cell culture supernatant was determined using HPLC analysis.And the results were shown that existence of 3 types of amino acid metabolism,that is,rapidly consuming amino acids(Ser?Gln?Arg?Val?Ile?Leu),slowly consuming amino acids(Thr?Cys?Tyr?Met?Lys?Phe)and accumulation of amino acids(Asp?Glu?Gly?Ala?Pro),and the amino acid metabolic type of 293 cells differs from other cells.(2)The changes of concentration of glucose and lactic acid in the culture medium were determined,and carbon source,energy metabolism and accumulation of lactic acid of 293 cells were analysed.And the results showed that glucose plays an important role in the growth and metabolism of 293 cells,and in the process of cell growth the content of glucose decreased and Lactic acid levels rises with the increase of culture time and cell number.(3)The growth of 293 cells was analyzed through cell counting and 293 cell growth curves.The maximum proliferation multiple of 293 cells per day was 1.95(doubling time was 24.62 hours).In most states,the proliferation multiple of 293 cells is lower than 1.95,indicating that the growth rate of 293 cells can be increased by adjusting the composition of the culture medium.(4)The concentrations of different amino acids and amino acids were analyzed on the effect of 293 cell culture.
