To characterize the different circulating miRNA expression in patients with AITD,we recruited forty-one patients who met criteria for GD,thirty-six patients of HT,and 41 healthy controls. The expression pattern of miRNAs in serum obtained from patients with GD and HT and healthy controls were determined by using Microarrays.Four interesting miRNAs(miR-155?miR-146?miR-21 ? miR-451)were selected andvalidated by TaqMan-based real-time PCR.And the relationships between the expression level of the above four miRNAs and the serum TSH level were analyzed by statistical methods. We found seventeen and sixteen miRNAs that were differently expressed in serum from patient with GD and HT respectively compared with healthy subjects by microarray analysis.Further qRT-PCR data consistently confirmed that serum level of miR-155?miR-146?miR-21andmiR-451 were increased in both GD and HT patients compared with healthy control. And the expression level of miR-155 was negativelycorrelated with the serum TSH level. Our results revealed that differential expression of serum miRNAs were exist in AITDs patients, and the expression level of miR-155 was associated with the function of thyroid, which may serve as biomarkers of AITD for diagnostic or treatment.
