Objective: To investigate the protective effect of ferulic acid on the brain of epileptic rat and its possible mechanism by analyzing the effects of ferulic acid on MDA,SOD,Bcl-2 and Bax in hippocampus of pentylenetetrazole(PTZ)-induced chronic epileptic rats.In order to provide clinical support for clinically better treatment of epilepsy.Methods:36 healthy male Wistar rats weighing at(240±20)grams were randomly divided into 3 groups: normal saline(NC)group(40 mg/kg daily intraperitoneal injection of saline solution)and pentylenetetrazole-induced epilepsy model(PTZ)group(40 mg/kg daily intraperitoneal injection of pentylenetetrazol)and ferulic acid(PTZ+FA)group(60 mg/kg intraperitoneal injection of ferulic acid 20 min before the injection of pentylenetetrazol daily)for 28 consecutive days.(1)Racine grades were recorded.(2)Changes in SOD activity and MDA content were detected.(3)the changes in the expression of Bcl-2 and Bax in the hippocampus were detected by immunohistochemistry.(4)The changes of Bcl-2 and Bax content in hippocampus were detected by Western Blot?Results: 1.Compared with PTZ group,Ratcine score in PTZ+FA group was significantly lower(P<0.01);Epilepsy latency was increased(P<0.01);SOD activity was significantly increased(P<0.01);The MDA content decreased significantly(P<0.01);Bcl-2 expression was significantly up-regulated in hippocampus(P<0.01),while Bax expression was significantly down-regulated(P<0.01).2.Compared with PTZ group,rats in NC group had significantly lower Racine score(P<0.01);SOD activity increased significantly(P<0.01);MDA content decreased significantly(P<0.01);Bcl-2 in tissue The expression was significantly increased(P<0.01),and the expression of Bax in the tissue was significantly reduced(P<0.01).Conclusion: 1.Ferulic acid has neuroprotective effect,and reduce seizures.2.Ferulic acid can reduce the rate of cell apoptosis through the anti-oxidative stress pathway,thereby protecting neurons.
