Objective: To investigate the expression and clinical significance of GSDMD(Gasdermin-D)in colorectal cancer tissues and adjacent normal tissues.Methods: The expression of GSDMD in cancer tissues and normal tissues was detected by immunohistochemical method and Western blot,and the relationship between the expression levels and clinical pathological data was explored.Results:1.General condition of the patient: 30 male patients and 22 female patients;patients aged 32-79 years,median age 55 years,age> 55 years,28 cases,age ? 55 years,24 cases;colon cancer patients,18 cases,34 cases of rectal cancer.Pathological classification: 43 cases of ulcers,9 cases of uplift.The degree of differentiation of tumor tissues was well-differentiated in 6 cases,moderately differentiated in 36 cases,and poorly differentiated in 10 cases.Lymph node metastasis: There were 16 cases without lymph node metastasis and 36 cases with lymph node metastasis.Tumor infiltration depth: T1 + T2 15 cases,T3 + T4 37 cases.Dukes stage: 16 cases in A+B period,36 cases in C period.The mucosal tissue 10 cm away from the same individual was used as a normal control.2.The expression of GSDMD in colorectal cancer tissues and normal tissues.The results of immunohistochemistry showed that the expression of GSDMD in colorectal cancer tissues was lower than that in normal tissues in 96% of patients(50/52)(P <0.05).The results of Western blot showed that the expression of GSDMD in colorectal cancer tissues was lower than that in normal tissues in 94%(49/52)patients(P <0.05).3.Relationship between GSDMD expression and clinical parameters in colorectal cancer.There was no significant correlation between the expression level of GSDMD and the age,sex,tumor pathological type and tumor location(P> 0.05).The expression level of GSDMD was statistically significant at the depth of tumor invasion,lymph node metastasis,tumor differentiation,and Dukes stage expression(P<0.05).The higher the expression level of GSDMD,the better the degree of differentiation of the tumor tissue,the lower the degree of infiltration,the earlier the clinical stage,and the smaller the probability of lymph node metastasis.Conclusions:1.The expression of GSDMD in colorectal cancer tissues is lower than that in normal tissues.2.The expression level of GSDMD in colorectal cancer tissues is significantly correlated with the degree of tumor progression.It may be used as an index to evaluate the occurrence,development and prognosis of colorectal cancer.
