Objective: 1、To identify the differential expression pattern of mi R-125b-5p in pancreatic cancer cells with different characteristics of invasion and metastasis.2、To clarify the function of mi R-125b-5p in regulating the morphology,invasion,metastasis and proliferation of pancreatic cancer cells.3、To identify the target gene of mi R-125b-5p.4、To investigate the impact of MAPK pathway on regulation of mi R-125b-5p expression.Methods: Realtime PCR(rt-PCR)was used to confirm the differential expression of mi R-125b-5p in pancreatic cancer cells with different characteristics of invasion and metastasis.mi R-125b-5p inhibitor was transiently transfected into highly invasive pancreatic cancer cells and mi R-125b-5p mimic was transiently transfected into poorly invasive pancreatic cancer cells.The morphological change of cells was observed.Wound healing assay,transwell assay and CCK-8 were used to clarify the function of mi R-125b-5p in regulating the morphology,invasion,metastasis and proliferation of pancreatic cancer cells.Bioinformatic method was used to predict the potential target genes of mi R-125b-5p and western blot was used to verify the target gene.Realtime PCR was used on mek2 knock down PC-1.0 cells to investigate the impact of MAPK pathway on regulation of mi R-125b-5p expression.Results: 1、mi R-125b-5p was upregulated in the highly invasive pancreatic cancer cells and was down-regulated in the poorly invasive pancreatic cancer cells.2、Overexpression of mi R-125b-5p increased pseudopodia of cancer cells and enhanced the invasion and metastasis while down-regulation of mi R-125b-5p resulted in inhibition of invasion,metastasis and proliferation of pancreatic cancer cells.3、SMAD4 was the potential target gene of mi R-125b-5p.4 、 Differential expression of mi R-125b-5p in invasive pancreatic cancer was regulated by MAPK signal pathway.Conclusions: 1、The expression of mi R-125b-5p is positively correlated to the invasion of pancreatic cancer.mi R-125b-5p promotes the invasion and metastasis of pancreatic cancer targeting SMAD4.2、The differential expression of mi R-125b-5p is regulated by MAPK signal pathway.
