Objective:We prepared conditioned media from culturing mouse BMSCs to investigate its effect on murine hair follicles,which may accelerate the hair cycle key stages from telogen to anagen.And to explore this effect whether it activate the process of Wnt signal pathways.Methods:The mouse BMSCs can be isolated and cultured by whole bone marrow culture.Conditioned media were collected at day 12 and day 15.All mice were divided into control group,test 1 group and test 2 group randomly to choose modeling optimization.Mice in test 1 group were day 12 CM injected from day 0 to day 10 on alternate days,while test 2 group were day 15 CM injected,and control group were media injected.The changes of hair cycle stages were verified by HE staining of paraffin sections at day 7,day 10,and day 14.The fluorescence quantitative PCR was used to detect the expression of molecules of Wnt signal pathways at day 7 and day 14.The data was analyzed with statistics method.Results:The skin color became black at day 6 in test 1 group.At day 14,there were 6 mice became black in test 1 group,while there were not any mice turned black in control group at day 14.At day 7,the transcript of?-catenin were increased in test 1 group.And at day 14,the expression ratio of?-catenin,Lef1,Tcf1 were significantly increased in test 1 group versus control group,while the transcript of other target genes were not different statistically between test group and control group.Conclusion:Injection of BMSCs-CM could accelerate telogen-to-anagen transition of murine hair cycle,significantly increase the expression of?-catenin,Lef1,Tcf1.And these are probably caused by the activation of Wnt/?-catenin signal pathway.
