Stress is the normal nonspecific physiological reaction when the body'was stressed,and it,promotes individual adaptability to adjust the environment.But strong stress makes individual life endangered,and make the body loses its normal adjustment function and unable to return to normality.post-traumatic stress disorder is always as the result of the stress After intense stress,the balance between the excitability of glutamate and the inhibitory of?-aminobutyric acid?GABA?in the brain can be destroyed,such as increased startle reflex,repeated frightening recall and negative state of high alert,which may be one of the molecular mechanisms of stress disorder.QYS can increase the expression level of GABAAR significantly to strengthen the action of the inhibiting system,and at the same time,reduce the excitability of glutamate to keep the balance between the excitement and inhibition.Therefore,QYS can affect the central nervous system and increase the avidity of GABA inhibitory effect,which maybe have a regulatory role in mental disorders induced by traumatic stress related with endocrine disorders of nervous system.First the chemical composition of the total glycosides of QYS has carried on the preliminary characterization by HPLC-MS in this paper,then PTSD model of fear condition successfully established,the system through the startle reflex and kuang field test PTSD induced enhancement of startle reflex,fear,anxiety behavior;At the same time by immune protein imprinting method to detect GABAAR expression changes.Finally,the use of qingyang and total glycosides different doses to explore qingyang and total glycosides on strong stress induced PTSD behavior and key brain regions GABAAR expression level adjustment.The results are as follows:According to the characteristics of the chemical composition of total glycosides types are mainly of two kinds of C21 steroids and acetophenone components,according to the standard quality of spectral behavior characteristics of the isolated,identified more than 33 components from total glycosides,including 4 kinds of acetophenone components derivatives and 29 kinds of C21 steroidal components.The component of C21 steroid is the dominant component of total glycosides.Model results show that compared with 2 d,14 d after shock stress can significantly induce rats fear behavior,startle reflex behavior.14 d can be used as PTSD model good behavior detection point in time.Stress after 14days,rat brain regions GABAAR a2,CeA,PL,DH?2 subtype expression levels drop;The medial prefrontal IL brain regions GABAAR a2 subtype expression level decreased,tip:brain regions can GABA inhibitory effect.QYS high-dose group significantly inhibited cues of the startle reflex induced enhancement and inhibit rats fear behavior,increase the activity of rats.Compared with model group,QYS group rats's CeA brain regions of amygdala?2 subtypes expression level inecreased.QYS group rats's DH brain regions of hippocampa GABAA receptor subtype expression did not change significantly,and the VH brain regions gamma 2 subtypes expression level increased.Conclusion:QYS may mainly through mediated CeA brain regions GABAAR?2 subtypes expression,to strengthen the function of inhibiting GABA system,with treatment of strong stress induced enhancement of fear and startle reflex behavior,and its active components were mainly C21Steroidal glycosides.
