Compound Prunella vulgaris is a clinical application in Wuhan Donghu hospital for many years.It is consists of six kinds of medicines like Prunella vulgaris,Rhynchophylla,Apocynum,Hawthorn,Cassiae Torae,Flos Daturae.It was used in the treatment of liver yang hyperactivity hypertension without side effect and with the functions of diuresis,lowering blood pressure,etc.A large number of study found that alkaloids compound in Prunella vulgaris has better antihypertensive activity.Establishing the high performance liquid chromatography ethod for the determination of the content of rhynchophylline under the guidance of the theory of traditional Chinese medicine,combined with the ideas and methods of modern development of Chinese medicine.Screening the extraction method of compound Prunella vulgaris,with rhynchophylline extraction rate as the index,compound Prunella vulgaris as a whole,on the basis of single factor experiment.The effects of liquid to solid ratio,ethanol concentration and soaking time optimal conditions.Using response surface method the optimum extraction process optimization.Study on the compound Prunella vulgaris infiltration percolation extraction and different polar parts of the preliminary separation,(I),chloroform and petroleum ether(II),ethyl acetate(III)were obtained,is butanol(IV)and ethanol(V)five parts.The animal experimental cats,antihypertensive substances test method for reference,antihypertensive activity of different polar parts were comparison,chloroform,petroleum ether and n-butanol parts has better antihypertensive activity,which antihypertensive effect best for chloroform,decompression amount greater than 4.0 kpa;Using column chromatography,the monomer compounds are isolated from the optimal activity of chloroform,and with the help of structural identification technology,the identification of compounds for rhynchophylline,scopolamine and dihydrogen celandine red base,to further explore the spica Prunellae compound antihypertensive activity component lay the foundation.Selecting rhynchophylline to study liposome based on the systematic study of the antihypertensive active ingredients of Prunella vulgaris.Liposomes were prepared by ethanol injection method of rhynchophylline,then Appearance characteristics,study of rhynchophylline liposome size distribution,encapsulation efficiency,leakage rate and in vitro release.Found that the liposomes in 48 h showed a relatively stable release effect.In this paper,the chemical compound of traditional Chinese medicine research and new formulations combine to better explain compound prunella spike of active substances and drop blood pressure mechanism,improve the effective components of the chemical stability and bioavailability.It has conducted the research on the preparation and curative effect of the compound Prunella vulgaris multi-target mechanism of action of inquiry and a new formulation of the research and development further use to provide the effective basis. |
