Objective: Using cross-linking agents to enhance the stiffness of dentin collagen and inactive the endogenous dentin proteases to prevent collagen degradation has been supported by many studies.This study was aimed to estimate the using of trimethyl chitosan quaternary ammonium salt(TMC)followed by1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC)on the properties change of dentin collagen.Material and methods: Human dentin was demineralized by 10% phosphoric acid for 48 hours,treated with 1mg/ml,5mg/ml and 10mg/ml TMC followed by 0.5M EDC.Specimens treated with deionized water as negative group and EDC as positive group.Then they were subjected to SEM and XPS test to see the morphological changes and the amount of TMC on the surface of collagen.Elastic modulus test,dry mass loss and HYP release was used to evaluate the mechanical properties and degradation.We use the CFU and CLSM to evaluate the antibacterial properties.Results: The use of TMC changed the morphology of collagen,with the increase in concentration,the amount of TMC attached to the surface of collagen increased.Significant differences were find in dry mass loss and HYP release and only high concentration of TMC+EDC can improve the elastic modulus,but all the cross-linking groups can reduce the decline.They also showed a significant antibacterial effect.Conclusion: The use of TMC with EDC on dentin collagen can increase the cross-linking,resistance to enzyme,mechanical property and antibacterial effect. |
