Objective: Expression of lysosomal-associated membrane protein-1(CD107a or LAMP-1)of natural killer cells was evaluated in untreated Epstein-Barr virus(E BV)DNA-positive patients.Combined detection with NK cell activity and perf orin supports a new way to evaluate changes of NK cell cytotoxicity after EB V infection,and provides a new selection of diagnosis and therapeutics.Methods: Peripheral blood mononuclear cells from the 39 patients detected pos itive EBV-DNA were isolated.Multi-parameter flow cytometry was used to det ected CD107 a levels of NK cells,and lactate dehydrogenase(LDH)release me thod was applied to analyzed activity of NK cells.14 healthy adults were eval uated as normal controls.Through inspecting clinical data and laboratory test r esults,data of Sorting-PCR,perforin expression and related gene detection were collected to figure out the relationship between NK cells and EBV infection.Results:1.Significant decrease of CD107 a expression on the surface of activat ed NK cells and activity of NK cells was observed 39 patients compared to c ontrols.And levels of perforin of NK cells were significantly lower in patients than controls,excepting for patients diagnosed as chronic active EBV infectio n(CAEBV).2.Compared to normal controls,patients with NK and T cells in fected by EBV exhibited a lower expression of CD107 a expression,activity of NK cells and perforin of NK cells.3.Of 5 patients detected with mutation o f NK cell degranulation related genes,three were detected less than 5% CD107 a expression on the surface of rest NK cells and four were observed reductio n of perforin expression in NK cells.Conclusion: Universal damaged NK cell cytotoxicity was discovered in EBV DNA-positive patients,with different levels as different target cells.It provides a new way to determine NK cell cytotoxicity and a new selection for diagnos is and therapeutics that combined detection with CD107 a expression,NK cell a ctivity and perforin.
