Alzheimer's disease(AD)is a common neurodegenerative disease that severely affects the patient's normal life.In general,patients suffering from AD characterized atrophy of brain,especially the atrophy of hippocampal in medial temporal lobes.At present,the pathogenesis and molecular mechanism of AD are still unclear,and there are no cures were clearly reported.Long non-coding RNA(lnc RNA)is a kind of non-coding RNA with more than 200 nucleotides.Studies have shown that lnc RNA has a variety of regulatory functions in the gene expression regulation.This study systematically investigated lncRNAs expressed in hippocampal tissues,identified novel lncRNAs that were differentially expressed between Alzheimer's disease and normal samples,and annotated novel lncRNAs by constructing lnc RNA-mRNA coexpression networks.The study mainly includes the following three aspects:(1)Identifying the lnc RNA in the case/control hippocampal body tissue.By de novo ab initio assembly of RNA-seq data and setting of screening conditions based on lnc RNA definition,1192 new lncRNAs that had never been annotated were identified.(2)Based on the case/control gene expression profile data,differentially expressed lncRNAs from hippocampal tissues in AD were obtained.(3)A coding-non-coding gene co-expression network was constructed by using 17581 coding genes and lncRNAs with FPKM ? 2 in 17 sample transcriptomes as the background set.The co-expressed genes of lnc RNA related to Alzheimer's disease suggested the molecular function of lnc RNA.GO functional annotation and KEGG pathway enrichment analysis showed that the differentially expressed lncRNAs were mainly enriched in such functional pathways as innate immunity,immune response,viral immunity,and cell adhesion.In summary,based on the RNA-seq data of hippocampal tissue in Alzheimer's disease patients,we first identified 1192 new lncRNAs in hippocampal tissues,and then performed differential expression analysis.11 up-regulated lncRNAs and 9 downregulated lncRNAs of the 1192 new lncRNAs were significantly differently expressed in AD.Finally,through the co-expression analysis of lnc RNA-mRNA coding gene,the lncRNAs in the co-expression network was annotated.This study is helpful for the understanding of the potential molecular mechanism of lncRNAs in AD and has important scientific significance.
