The Expression And Function Analysis Of YRDC In Lung Cancer

Studying the expression and function of genes in cancer can help us understand the impact of cancer genes and the mechanisms of cancer development.It is of great significance to guide cancer treatment.In the study of cancer gene c1orf109 in human chromosome 1p34.1,found that genes in this region can express together.YRDC(named yrd C N6-threonylcarbamoyl-transferase domain containing)is one of these genes in this region which predicted to be highly expressed in cancer,but it's biology function is unclear.Based on the previous studies,this study focused on the differences in the expression of YRDC in tumors and the effects of YRDC on the biological function of tumors,explored the possible molecular mechanisms of YRDC.First,using bioinformatics combined with TCGA database,we analyzed the expression differences of YRDC in lung cancer,the relationship of YRDC expression with cancer stage,gender,age and smoking habits and the YRDC expression in some lung cancer cell lines.Second,the YRDC-knockdown and the YRDC-overexpression cell lines were constructed to test the ability on cell proliferation,cell migration and cell invasion.Finally,the interaction proteins were predicted with bioinformatic and the Prokaryotic expression plasmid was constructed to purify the YRDC protein for verifying the biology function of YRDC.The results showed that the YRDC gene showed a high expression tendency in lung cancer,and there was no significant difference in stage,gender,age and smoking habits.The cell proliferation,migration,and invasion were repressed when YRDC was down-regulated.Overexpression of YRDC gene promoted the cell proliferation,migration,and invasion.Interaction protein prediction showed there are many interactive molecules with YRDC.YRDC homologous structure displayed that the structures of YRDC and E.coli are not highly similar and their function maybe different.Protein activity test showed that the YRDC has the ATP Enzyme activity which required for t RNA modification and the activity was more effective compared to YRDC in E.coli.In summary,this article explores the YRDC gene expression in lung cancer,confirmed that the YRDC gene has a certain significance for tumor development,YRDC gene expression will reduce the physiological activity of cancer cells,human YRDC gene may still have t RNA modification activity.Its molecular mechanism is still related to the translation of proteins.
