Objective: To explore the association between rs174547 and rs174547 loci in FADS1(Fatty acid desaturase 1)gene and rs1535 locus polymorphism in FADS2(Fatty acid desaturase 1)gene and Head and neck squamous cell carcinoma(HNSCC),in order to provide theoretical evidences for the etiological study and scientific prevention and treatment of HNSCC.Methods: A case-control design was used to study the association between the selected SNPs and HNSCC,including 515 HNSCC cases in otolaryngology and otolaryngology department of Tongji Hospital and Union Hospital of Wuhan City and 515 controls according to the 1:1 matching principle of sex and age(±5 years old).Collect each subject 2ml venous blood,basic demographic information as well,the basic information of the cases collected by medical records,and information of controls by questionnaire.After DNA extraction,rs174547 and rs174547 loci of FADS1 genes and rs1535 loci of FADS2 gene were genotyped by im LDR multiplex SNP typing technique.The relationship between rs174547 rs174547 and rs1535 polymorphism loci and HNSCC was analyzed by Chi-square test and logistic regression analysis using SPSS 20.0.Results: The genotypic distribution of rs174547 rs174549 and rs1535 in the control population met the Hardy-Weinberg equilibrium.Multivariate logistic regression analysis showed that rs174547 TT and CT genotypes were 1.70 times(95%CI=1.21-2.41,P<0.01)and 2.15 times(95%CI=1.47-3.14,P<0.01)more likely to occur than CC genotypes.Compared with the rs174549 GG genotype,AA genotype reduced the risk of HNSCC by 0.45 times(95%CI=0.31-0.65,P<0.01),and GA genotype showed no significant difference(P = 0.11).Compared with rs1535 AA genotype,GG genotype reduced the risk of HNSCC by 0.46 times(95%CI=0.32-0.67,P<0.01),and GA genotype showed no significant difference(P = 0.08).Combined effect analysis showed that compared with individuals not carrying candidate SNP risk alleles,individuals with 1-2 risk alleles and 3 risk alleles were 1.71(95%CI=1.22-2.40,P=0.002)and 2.24 times(95%CI=1.54-3.26,P<0.001)more likely to develop HNSCC,respectively.Interaction analysis showed that the risk of HNSCC in smokers was higher than in non-smokers.Conclusions: In this study,we found that there was a statistically significant association between the polymorphic loci rs174547、rs174549 on FADS1 gene and rs1535 on FADS2 gene,which need to be verified by functional studies. |
