Objective:To hepatocellular carcinoma in situ?Hepatocellular carcinoma HCC?mouse model to observe the effect of electroacupuncture adjusting effect on liver cancer in mice with spontaneous wheel wheel activity rhythm of epigenetic mechanisms.Methods:After the 108 male C57BL/6J mice were strictly selected according to6 zeitgeber time?Zeitbeger Time,ZT?were randomly divided into ZT0 group?07:00?,ZT4?11:00?group,ZT8 group,ZT12?15:00??19:00?group,ZT16?23:00?group and ZT20?03:00?group.Each group was respectively set blank control group,model group and EA group.Male C57BL/6J mice control group amplitude,the differences were statistically significant?P<0.05?;the animal model group the peak value and phase respectively before and after modeling,compared with the control group the difference of peak phase difference compared with the control group,the difference of amplitude,the differences were statistically significant?P<0.05?;but the animal model group starting time,and cycle respectively before modeling,compared with the control group the difference between before and after the animal activity starting time,cycle the value of difference comparison,the differences were not statistically significant?P>0.05?.?After acupuncture,orthotopic transplantation of mice in the EA groups the activity rhythm of the runner is reduced and the peak amplitude increased lag ZT8 EA orthotopic transplantation mice wheel activity rhythm is the most effective performance for the ZT8 EA group can significantly increase the amplitude of mice decreased runner activity rhythm,the peak phase advance lag.The electroacupuncture group activities in mice starting time,and cycle respectively before and after acupuncture,difference with the model group MICE activities starting time,cycle difference compared with the control group,MICE activities starting time,cycle difference comparison,the differences were not statistically significant?P>0.05?.?After modeling,the mice in ZT8 model group SCN 84 epigenetic related genes in which 33 upregulated and 6 genes related to DNA methylation,histone acetylation related gene 13;histone methylation related gene 14.?After acupuncture,electroacupuncture group ZT8 mice SCN 84 epigenetic gene31 expression,including 5 genes related to DNA methylation,histone acetylation related gene 11;histone methylation related gene 15.Conclusion:?The normal mice spontaneous activities showed the runner circadian rhythm characteristics.?The successful establishment of orthotopic liver cancer mouse model,the spontaneous activity rhythm of runner showing reduced amplitude,peak phase lag phase characteristics.?The spontaneous activity rhythm after acupuncture,the runner performance orthotopic transplantation mice:the peak phase amplitude increased,reduce the forward backward,the adjustment function in ZT8 time point electroacupuncture on orthotopic transplantation of mice spontaneous wheel activity rhythm of the most significant performance for the amplitude increased most significantly,only ZT8 time point can peak phase forward backward.?The effect of Electroacupuncture on orthotopic transplantation of mice with SCN liver cancer in view of genetic genes related to certain tables,such as DNA methyltransferase 3B?Dnmt3b?,histone deacetylase 3?Hdac3?,histonelysine methyltran ferase 1a?Kdm1a?,histonely sinede methylase 2e?Kmt2e?gene downregulation,may participate in the adjustment of EA orthotopic transplantation mice spontaneous wheel activity rhythm within the SCN epigenetic molecular mechanism.
