| Objective:This study is based on the traditional Chinese medicine "the spleen and pancreas with homologous" thought "yangyinyiqihuoxue efficacy of Shenqi compound used in GK animal model of diabetic macrovascular disease in rats and study its effect on the general condition,blood glucose,blood lipid,CRP,aortic and pancreatic pathology in GK rats,and through the daily average blood glucose levels(MBG)the average daily blood glucose,standard deviation(SDBG),maximum blood glucose fluctuations(LABG)and serum GLP-1,preliminary study of Shenqi compound with Yangyin Yiqi Huoxue method can reduce blood glucose fluctuation,so as to achieve the purpose of intervention of diabetic macrovascular disease.Methods:40 male GK rats were selected and the blood glucose was measured by strict blood glucose monitoring.According to the blood glucose level,the blood sugar was numbered from high to low,and 10 of the random blood sugar were less than 11.1mmol.L-1.In the end,30 rats were conformed to the requirements,and 30 male GK rats of 30 2 times of blood glucose or more than 11.1mmol.L-1 were randomly divided into the male rats.The 3 groups: blank group(n=10),Shenqi compound group(referred to Shenqi group,n=10),Western glipetine group(n=10),and 10 5 month old male Wistar rats as normal control group(abbreviated normal group,n=10),total of 4 groups.After 4 weeks of adaptive feeding,the blank group was given ordinary feed and sterilized pure water,the Shenqi group,the model group,and the group of Western glipetine were given high fat diet,and the high fat diet was given continuously,and the diabetic macrovascular disease animal model was established.Then the blank group and the model group were treated with 5ml/kg.D,and Shenqi group was filled with Shenqi compound extract according to 1.44g/kg.D,while the group of Western glipetine was given Sig Leo Dean d with 16mg/kg.D for 8 weeks.The general condition of the animals was observed during the experiment.During the fourth week of gavage,the blood glucose levels of 8:00,10:00,14:00,16:00,18:00 and 5 time intervals were measured once a week,and the values of MBG,SDBG and LABG were calculated.GK rats were killed after 8 weeks of gavage.Blood lipid level,CRP,serum insulin,serum glucagon and serum GLP-1 were detected in each group.The abdominal aorta and pancreatic tissue were stained with HE to observe the pathomorphologyResults:The general condition of Shenqi compound group is better than that of the model group.The average blood glucose standard deviation(SDBG)and the maximum blood sugar fluctuation range(LAGE)of blood glucose fluctuation are better than those of the model group.Triglyceride(TG),cholesterol(TC)and C reactive protein(CRP)were lower than those in the model group,but no statistical significance was found in the western medicine group.There was no significant difference in insulin(INS)and glucagon(GC)compared with the model group,but GLP-1 was significantly different from that in the model group,P < 0.01.In pathology,the thoracic and abdominal aorta slices of Shenqi compound group showed complete smooth endometrium,middle membrane elastic fibers,and cell nuclei arranged regularly.No islets were found in the pancreas slices,most of the nuclei were pyknosis and apoptosis,and there was no abnormal change in the size and shape of the islets.Conclusion:The general state of the application of Shenqi compound recipe can improve the vascular lesion in GK diabetic rats,increase GLP-1 concentration,fluctuation of blood glucose control;and can improve lipid metabolism,decrease CRP and alleviate vascular endothelial injury in rats model of GK,delay the progress of diabetic macrovascular disease. |
