The Inhibitory Effect Of Cd8?~- Dendritic Cells From Soluble Egg Antigen Immunized Mice On OVA-induced Allergic Asthma

Objective:The purpose of this research is to grope for the inhibitory effect of dendritic cells(DCs)from Schistosoma japonicum-soluble egg antigen(SEA)infected mice on OVA-induced allergic asthma and its mechanism.Method:We chose 5 PBS control mice and 5 SEA infected mice to extract the spleen and prepare for CD8?~+DCs and CD8?~-DCs by MACS.The different DCs from PBS-control mice and SEA-treated mice were tagged as PBS-CD8?~+DCs,PBS-CD8?~-DC,SEA-CD8?~+DCs and SEA-CD8?~-DCs respectively.We chose another 30 BALB/c female mice to divide into 6 groups randomly:Normal group,only OVA group,PBS-CD8?~+DCs/OVA group,PBS-CD8?~-DCs/OVA group,SEA-CD8?~+DCs/OVA group,SEA-CD8?~-DCs/OVA group.PBS-CD8?~+DCs and PBS-CD8?~-DCs were respectively transferred to PBS-CD8?~+DCs/OVA group and PBS-CD8?~-DCs/OVA group.SEA-CD8?~+DCs and SEA-CD8?~-DCs were respectively transferred to SEA-CD8?~+DCs/OVA group and SEA-CD8?~-DCs/OVA group.Then,the allergic asthma was induced by OVA synchronously except Normal group.Mice of Normal group were used as control.1.Lung histopathological examination of mice by H&E staining.2.Collect Bronchial alveolar lavage fluid(BALF)from different groups and determine IL-4,IL-5,IL-10 and TGF-?protein levels by ELISA.3.Splenocytes were cultured in 6-well plate and stimulated with OVA for 72h.Then collect the Splenocytes culture supernatant(SCS)from all groups to confirm the expression levels of IL-4,IL-5,IL-10 and TGF-?by ELISA.4.Collect blood and prepare serum for measurement of OVA-specific IgE by ELISA.Result:1.Compared to that of only OVA group,PBS-CD8?~+DCs/OVA group,PBS-CD8?~-DCs/OVA group,SEA-CD8?~+DCs/OVA group,the lung inflammatory responses in SEA-CD8?~-DCs/OVA group was alleviated,and decrease of inflammatory cells infiltration and bronchial mucus secretion.And,pathological score of lungs from SEA-CD8?~-/OVA group was 6.80±1.30,lower than only OVA group and other adoptively transferred groups,which were 14.00±1.00,13.00±0.82,12.75±0.50,and 11.00±0.00 respectively(P<0.05).2.IL-4 and IL-5 levels of BALF and SCS from SEA-CD8?~-DCs/OVA group were lower than that of only OVA group and other adoptively transferred groups(P<0.05).3.IL-10 and TGF-?levels of BALF and SCS from SEA-CD8?~-DCs/OVA group were higher than that of only OVA group and other adoptively transferred groups(P<0.05).4.Level of OVA-specific IgE from SEA-CD8?~-DCs/OVA group were lower than that of only OVA group and other adoptively transferred groups(P<0.05).Conclusion:1.CD8?~-DCs from SEA-immuned mice inhibit the allergic asthma induced by OVA.2.Adoptive immunity of SEA-CD8?~-DCs inhibit asthma by reducing IL-4,IL-5production.3.Adoptive immunity of SEA-CD8?~-DCs inhibit asthma by augumenting IL-10,TGF-?production.
