| Objective In this paper,a new type of adjuvant for biodegradable polymer microspheres was studied.Due to its advantages of controllable particle size,slow release of antigens,targeting,and effective enhancement of humoral cellular immune responses,it has become one of the focused areas for the development of new adjuvants in recent years.Methods In this paper,the particle size and dispersion of chitosan microspheres were investigated by a single gel formation method.The screening of protective agents during the subsequent spray drying process was investigated.Dry microspheres were made into dry powders.At the same time,the application of chitosan glutamate and pickering emulsion was investigated.The mouse was used as an animal model to evaluate the effect of different adjuvants and antigen binding.Results The microspheres were successfully prepared with a uniform and well-dispersed particle size of approximately 1 μm,the optimal concentration of the protective agent was selected to maintain the integrity and the original performance of the microspheres during spray drying.At the same time,chitosan gel microspheres can be applied to chitosan salts and pickering emulsions of other materials,and good immune evaluation can be obtained.Conclusions The study found that the chitosan gel microspheres prepared by the single-phase gelation method are uniform in particle size,well dispersed,and can be repeatedly prepared.The addition of the protective agent after spray drying makes the microsphere vaccine have a higher level of immune response,making it safe,effective,easy to transport,easy to store and so on. |
