Objective: To observe the expression of GRP78,CHOP,Bcl-2 and Bax proteins in the control group and low-grade and high-grade gliomas,and to investigate the role and mechanism of endoplasmic reticulum stress CHOP pathway on apoptosis of glioma.Methods: We collected 30 cases of glioma patients and 5 cases of normal brain tissue from neurosurgery in our hospital in 2017.The tumor tissues were pathologically graded by our department of pathology,and were classified as low-grade glioma group(WHO I-II)in 14 cases and high-grade glioma group(WHO III-IV)in 16 cases.Normal brain tissue 5 Cases belong to the control group.A part of the specimen was packed into a block of wax,and apoptosis was detected by TUNEL assay;the other part was stored in liquid nitrogen and Western blot was used to detect the levels of GRP78,CHOP,Bcl-2,and Bax protein.Results: TUNEL assay showed that there were more apoptotic cells in the glioma group than in the control group,and the apoptotic index was high.The low-grade glioma group had more apoptotic cells and higher apoptotic index than the high-grade glioma group.Western Blot showed that GRP78,CHOP,Bcl-2,and Bax protein expression in glioma group increased compared with control group;GRP78,CHOP and Bax protein expression in high-grade glioma group was higher than in low-grade glioma group.Bcl-2 protein expression is low.Conclusions:1.In the low-grade glioma group,there was more apoptosis in the higher-grade glioma group and the apoptotic index was increased,indicating that apoptosis was negatively correlated with the degree of malignancy of the glioma.2.The expression of GRP78 protein in glioma group was significantly higher than that in control group,indicating that glioma initiates the endoplasmic reticulum stress apoptotic pathway;the increase of GRP78 protein is negatively correlated with the degree of malignancy of glioma,indicating that high-grade glial There may be a mechanism of apoptosis in the tumor against endoplasmic reticulum stress.3.Apoptotic index increased in glioma group,and GRP78,CHOP,Bcl-2and Bax protein expression increased,indicating that endoplasmic reticulum CHOP pathway is involved in the apoptosis of glioma cells.4.Activation of endoplasmic reticulum stress CHOP pathway promotes apoptosis,may be a new idea for the treatment of glioma.
