Objective:A MPE mouse model was created by injected with LLC through pleural,and intervened with anti-IL-9 antibody.To observe the expression level and learn its interrelation,we tested the expression of Th9cell,Th17 cell,Th9 cell,IL-17,IL-9,ROR?t and PU.1 in peripheral blood?MPE and spleen.Methods:Wild type?WT?female C57bL/6 mouse and IL-10 knock out mouse(IL-10-/-)with the same background were selected.Random number table method was used,then two type mouse were respectively divided into two groups,and random selected one group from different type mouse,then intervened with anti-IL-9 antibody.four groups:WT,WT+anti-IL-9,IL-10-/-,IL-10-/-+anti-IL-9.We examined the expression level of CD34 and Ki67 in pleural tumors tissue with immunohistochemistry method;The expression of Th17 cells and Th9 cells were detected in peripheral blood,MPE and spleen with Flow cytometry;we tested the expression level of il17,il9,ROR?t,PU.1in spleen with RT-PCR technology;we also gauged The level of IL-17 and IL-9 in peripheral peripheral blood and MPE with ELISA.Results:1.Compared with WT group,s CD34 and Ki67 in pleural tumor,WT+anti-IL-9groupwasincreased,whileIL-10-/-groupand IL-10-/-+anti-IL-9 group was decreased,and IL-10-/-+antiIL-9 group was more decreased than IL-10-/-group.2.Compared with WT group,s Th17cell,il-17,ROR?t in peripheral blood,MPE and spleen with Flow Cytometry?ELISA?RT-PCR methods,WT+anti-IL-9 group was decreased in different degrees;IL-10-/-group was increased in different degrees;IL-10-/-+anti-IL-9 was increase in different degrees,and higher than IL-10-/-+anti-IL-9 group.3.Compared with WT group,s Th9 cell,IL-9,PU.1 in peripheral blood,MPE and spleen group with Flow Cytometry?ELISA?RT-PCR methods,WT+anti-IL-9 group,IL-10-/-group and IL-10-/-+anti-IL-9 group all decreased in different degrees,the expression leavel from high to low was WT+anti-IL-9group,IL-10 knock out group,10 knock out+anti-IL-9 group.Conclusions:1.Antagonisming the effect of IL-9,can reduce the expression of IL-17?ROR?,elevate the expression of CD34 and Ki67,promote the formation of new blood vessels and proliferation of tumor cells.2.Knocked out IL-10,can elevate the expression of IL-17,reduce the expression of IL-9?PU.1,reduce the expression of CD34 and Ki67,inhibite the formation of new blood vessels and proliferation of tumor cells.3.removed the effect of IL-9 and IL-10 at the same time,can elevate the expression of IL-17,reduce the expression of IL-9,reduce the expression of CD34 and Ki67,inhibite the formation of new blood vessels and proliferation of tumor cells,and those effects were stronger than remove IL-10only.
