Objective: To explore the prevalence of sleep disorder, the main symptomatic characteristics and its association between cognitive function in the patients with Alzheimer's disease. And further estimate the value of Memantine in the sleep disorder of Alzheimer disease. Methods: We recruited 32 patients with Alzheimer's disease from neurology outpatient of First Hospital Affiliated to China Medical University during Feburary 2016 and October 2017 with MMSE score ranging from 10 to 20.After obtaining the informed consent of the subjects, general information,MMSE,HAMD,PSQI and ESS score was collected by professionally trained neirologists.A total of 32 subjects were followed up by telephone in December 2017,and 24 subjects were successfully followed up,with 8 subjects lost their follow-up.The subjects were asked in detail about sleep conditions and medication, and was evaluated by the PSQI scale again. The Epidata3.0 software were used to establish a database of collected data. After proofreading,SPSS 22.0 statistical software were used for analysis. The prevalence of sleep disorders in AD patients,PSQI subproject distribution of AD patients with sleep disorders.the correlation between MMSE and PSQI and ESS in AD patients, the impact of Memantine on sleep condition in AD patients were calculated respectively.Results:(1)The prevalence of sleep disorders in 24 AD patients was 50%.Among the 12 AD patients with sleep disorder, the percentage of PSQI in descending order were: sleep efficiency(66.7%),sleep quality(58.3%),sleep duration and sleep disturbance(both 50.0%),daytime dysfunction(42%)and total sleep time(41.2%).(2)The Spearman correlation coefficient of MMSE and PSQI was-0.656,with difference statistically significant(P < 0.01),and the Spearman correlation coefficient of MMSE and ESS was 0.159,with difference not statistically significant(P > 0.05).The results indicates that the severity of sleep disorder in AD patients is negatively correlated with cognitive function.(3)The total sleep duration after treatment of Memantine was statistically significant(P < 0.05).There was no statistical difference in PSQI total score and other subprojects compared to pre-medication(P > 0.05).Conclusion:(1)The most significant reduction was in sleep efficiency and sleep quality among AD patients with sleep disorders.(2)The sleep disorders and cognitive functions of AD patients interact with each other, and they are closely related.(3)The Memantine can extend the total sleep time of AD patients.
