Objective:Part one:The goal was to explore the relationship between Galectin-9-Tim-3and CRS by analyzing of the differences in the expression of Galectin-9 and Tim-3 in nasal mucosa of CRS patients and the control group.Part two:The purpose of this experiment was to study the expression intensity and distribution of Galectin-9 and its receptor T cell immunoglobulin domain mucin domain protein-3(Tim-3)in nasal mucosa of sinusitis rats from m RNA and protein levels by establishing rat rhinosinusitis model,to analysis of the role of Galectin-9 and Tim-3 in the pathogenesis of nasosinusitis.Methods: Part one:Selected patients with CRS who were hospitalized in our department from September 2016 to December 2016 as experimental group(group A).Patients with nasal septum were selected in our hospital with nasal as the blank control group(group B).All of the above subjects took the mucosa of the sinus oral and nasal complex as an experimental specimen during the operation.Part two: 30 SD rats were randomly divided into 3 groups,group ROS(group C),group CRS(group D)and blank group(group E).In group C and D,the CRS model was established by Streptococcus pneumoniae plus nasal filling.After 28 days of modeling,the group C was given with ROS for 7 days,group D and E group were intragastrically administered with normal saline.The sinuses of the three groups were taken as specimens.All animals and clinical specimens were detected by real-time fluorescence quantitative PCR(q PCR),hematoxylin-eosin staining(HE staining)and immunohistochemistry,to detect the distribution and expression of Galectin-9 and Tim-3in the nasal mucosa of each group.The SPSS17.0 software was used to analyze the data of each group and to compare the differences between each group.To discussed the relationship between the Galectin-9/Tim-3 and the incidence of CRS.Results:Part one:(1)Galectin-9 and Tim-3 were expressed in the nasal mucosa of group A and group B,and the expression of Galectin-9 and Tim-3 in A group was significantly higher than that in group B,and the difference was statistically significant(p<0.05).(2)Pearson correlation test and linear regression graph found that the linear correlation coefficient of Galectin-9 and Tim-3 m RNA and protein expression in group A and B were r>0 and p<0.05,suggesting that the expression of Galectin-9 and Tim-3 was significantly correlated and positively correlated.Part two:(1)Galectin-9 and Tim-3 were expressed in the nasal mucosa of rats in group C,D and E.the expression of group C and D were higher than that of E group.the difference were statistically significant(P<0.05).After ROS intervention,Galectin-9 and Tim-3 expression in group C and group D showed no significant difference,and the difference was not statistically significant(P>0.05).(2)Pearson correlation test and linear regression graph found that the linear correlation coefficient of the expression of Galectin-9 and Tim-3 m RNA and protein in the three groups was r>0 and p<0.05,suggesting that the expression of Galectin-9 and Tim-3 was significantly correlated and positively correlated.Conclusion:(1)Galectin-9 and Tim-3 are related to the inflammatory process of sinusitis,and play an inflammatory regulatory role,which may be involved in the development of sinusitis.(2)ROS may have no interference effect on the expression of Galectin-9 and Tim-3in the rat rhinosinusitis model constructed by Streptococcus pneumoniae with nasal packing method.
