Objective: The aim of this study was to investigate the expression of homeodomain-Interacting Protein Kinase 2(HIPK-2)in various molecularly-differentiated breast cancer tissues and to investigate the ?-catenin of the signaling pathway of WNT / ?-catenin in various molecular breast cancer tissues. To further explore whether HIPK-2 through WNT / ?-catenin signaling pathway affect the proliferation, invasion and metastasis of triple-negative breast cancer lay the foundation. Methods: The study selected 12 cases of breast cancer patients with tumor tissue and normal tissues(luminal A type in 3 cases, luminal B type in 3 cases,HER-2 type in 3cases,triple-negative type in 3 cases).Detecting the protein expression levels of HIPK-2and ?-catenin in various molecularly-differentiated breast cancer tissues and their normal tissues by Western blot.Statistical analysis the differences of HIPK-2 and ?-catenin protein relative expression in various molecularly-differentiated breast cancer tissues and their normal tissues;analysis the differences of HIPK-2 and ?-catenin protein relative expression in triple-negative breast cancer and the other three types of breast cancer;analysis the correlation between HIPK-2 and ?-catenin protein expression in 12 cases of breast cancer. Results: The results of Western blot showed that : in various molecularly-differentiated breast cancer, the expression of HIPK-2 in cancer tissues was lower than that in the same type of normal tissues the difference was statistically significant(P <0.05),The expression of ?-catenin in cancer tissues was higher than that in the same type of normal tissues, the difference was statistically significant(P <0.05).The expression of HIPK-2 in triple-negative breast cancer tissues was relatively lower in all molecular breast cancer tissues the difference was statistically significant(P <0.05),while the expression of?-catenin in triple-negative breast cancer tissues was relatively Higher, the difference was statistically significant(P <0.05).In breast cancer tissues, the relative expression of HIPK-2 and ?-catenin protein showed a negative correlation, the correlation coefficient r=-0.672.Conclusion: In various molecularly-differentiated breast cancer,HIPK-2 expression level in tumor tissue is lower than that in the same type of normal tissue, and ?-catenin is higher in tumor tissue than in the same type of normal tissue. In various molecularly-differentiated cancer tissues,HIPK-2 was the lowest expression in the triple-negative breast cancer tissues and ?-catenin was highest in the triple-negative breast cancer tissues. The expression of HIPK-2 and ?-catenin in breast cancer tissues has a negative correlation, and the effect of HIPK-2 on the proliferation, invasion and metastasis of breast cancer may be related to the WNT/?-catenin signaling pathway.
