Objective: Isolation and identification of actinomycetes from the special habitats in the northern part of the Guizhou Province and identification of potential strains and new strains to provide genetic resources and actinomycetes species for the development of new antibiotics and active secondary metabolites;Streptomyces isolated from Guizhou,a typical habitat for the source of Streptomyces sp.FJS 31-2.The secondary metabolites of the Streptomyces sp.FJS 31-2 strain were fermented,isolated and purified to obtain novel active secondary metabolites of structure.Methods: collecting soil samples in the undisturbed special habitats of north Guizhou such as Fenghuang Mountain,Da Banshui,Quaternary Ditch,and Buddha Light Rock;using the GYM culture medium and water AGAR and ISP2 to separate and purify single colony;testing the activity of the fermentation products and narrowing the range of the active site with the help of high performance liquid phase;separating the target compounds by large-scale fermentation,extraction,positive-phase silica gel column chromatography,sephadex LH-20 glucan gel column chromatography and high performance liquid chromatography.Results: 318 isolates are isolated and purified,and a group of strains with good anti-MRSA activity are selected.In addition,12 possible new strains are found by evolution analysis of 16 s RNA system,and Gene screening and resistance test are carried out on more than 99 active strains.Two monomer compounds are isolated from Streptomyces sp.FJS 31-2.Conclusion: it can be found that 36.16% of the isolates strains are resistant to MRSA activity through 16 S rRNA comparison after amplification ratio and phylogenetic tree analysis,which shows strains resources of resistant MRSA activityhave great development potential in north Guizhou and Streptomyces sp.FJS 31-2 can produce new halogenated natural products,which has important medical value.
