Part one:The strategy of postpartum depression model building[Objective]The present experiment is designed to explore the preferring period of CORT injection to build the postpartum depression model,which is vital to elucidate the possible mechanism of postpartum depression and study the effect of anti-postpartum drug.[Methods]Firstly,male and female mice was crossed together,pregnancy was determined by the vaginal suppository,Secondly,adult female ICR mice were randomly divided into six groups depending on the period of CORT/Oil injection,the preferring period is selected as followings:Day 1-Day 10 of pregnancy;Day 1-Day 21 of pregnancy;Day 10-Day 21 of pregnancy;Day 1 of pregnancy to Day 21 of postpartum;Day 10 of pregnancy to Day 21 of postpartum;Day 1 to Day 21 of postpartum,each group were further divided into experimental group(CORT injection)and control group(Oil injection)at random,The model group were injected with corticosterone to build postpartum depression,while the control group were treated with oil.Behavioral experiments including open field test(OFT),elevated plus maze test(EPM),forced swimming test(FST)and tail suspension test(TST)were performed to check the preferring period of CORT injection to build postpartum depression model.[Results]The immobility time significantly increase after CORT injection during G1-G21?G1-P21?G10-G21 and G10-P21 as compared to oil injected group in the FST,also the immobility time significantly increased after CORT injection during G1-G21?G1-P21 and G10-P21 compared with oil injected group in the TST.The frequency traveled in the open arm of the mice,who is injected with CORT during G10-G21,significantly increased compared to oil treated group in EPM;While,the frequency traveled in the open arm of the mice,who is injected with CORT during G1-P21,significantly decreased compared to oil treated group in EPM.The total distance traveled and the frequencies in the centre zone of the mice,who is injected with CORT during G1-G10,significantly increased compared to oil treated group in OFT;The total distance traveled of the mice,who is injected with CORT during G10-G21,significantly increased compared to oil treated group in OFT;While,the frequencies in the centre zone of the mice,who is injected with CORT during P1-P21,significantly decreased compared to oil treated group in OFT.According to experimental statistics,injected with CORT during pre-pregnancy(G1-G10)can result in higher miscarriage rates and maternal mortality.[Conclusions]Continuous hypodermic injection of corticosterone during the four periods:G1-G21?G1-P21?G10-G21 and G10-P21 can induce the depression-like behavior.But the pre-pregnancy(G1-G10)can result in higher miscarriage rates and maternal mortality,while,the periods:G10-G21 and G10-P21 can be used as a preferring period to build postpartum depression model.Continuous hypodermic injection of corticosterone during the P1-P21 can induce the anxiety behavior,which can be used as a preferring period to build postpartum anxiety model.Part two:the antidepressant effects of resveratrol on cognitive dysfunction[Objective]The current study was designed to examine the antidepressant effects of resveratrol(RSV),also its possible mechanism specialized on improving cognitive function.[Methods]C57BL/6J mice was separated four group,control,Model,Model + RSV,and Model + NA + RSV,all mice subjected corticosterone(20 mg/kg/d)intraperitoneal injection for 21 consecutive days except the control mice.From the 22nd to 42nd days different group received further treatment with vehicle/RSV(400 mg/kg/d.op)/NA(100 mg/kg/d.ip)+ RSV(400 mg/kg/d.op).The classical behavioral tests,the sugar preference test,the novel object learning test,the novel place learning test and the water maze test were applied to evaluate the acknowledgement effects of RSV.Subsequently,the PGC-1??SIRT1 and PV gene transcription and translation level were evaluated by RT-PCR and western blotting.[Results]The depression model mediated by CORT injection is evaluated by the sugar preference test.The mice showed significant increased proration on the frequency?duration and distance in the novel object learning test following the RSV administration,significant decreased proration on the time and distance to find the platform in the water maze test.All the results are accompanied with the increase in protein expression of SIRT1,and PV,while the cognitive dysfunction cannot be reverse with the SIRT1 inhibition-nicotinamide injected head of the RSV administration.[Conclusions]Resveratrol can reverse the cognitive dysfunction of depressant mice,which may be achieved by activating the SIRT1/PGC-1? signaling pathway and increasing the transcription and protein expression of PV.
