Objective: To examine the expression pattern of Annexin A1 in glioblastoma's clinical samples,para-carcinoma tissues and cell lines.To investigate the effect of Annexin A1 on glioblastoma's proliferation,migration ability,invasiveness,apoptosis and radiosensitivity.To explore the underlying molecular mechanism by which Annexin A1 mediates glioblastoma's biological behavior.Methods: Western botting and qRT-PCR were employed to examine the expression levels of Annexin A1 in glioblastoma's clinical samples,para-carcinoma tissues and glioma cell lines.We then constructed lentivirus vector that targeted human Annexin A1 and the negative control lentivirus vector and transfected them into glioma cell lines U87 and U251.Cck-8 assay,colony formation assay,Scratch wound healing assay,chamber invasion assay and flow cytometry were applied to evaluate glioma cell lines' proliferation,migration ability,invasiveness,apoptosis and radiosensitivity respectively.We further probed the differential expressed molecules between Annexin A1-knockdown group and negative control group by using Western blotting,qRT-PCR and enzyme-linked immunosorbent assay.Results: Annexin A1 was highly expressed in glioblastoma clinical samples and glioma cell lines U87 and U251,but not in para-carcinoma tissues.Knocking down Annexin A1 suppressed glioma cell lines' proliferation,migration ability,invasiveness and sensitized them to radiation.We also found out that Annexin A1 knockdown down-regulated the expression levels of interleukin 8 and ihhibited the phosphorylation levels of p65,which is a key member of NF?b family.Conclusion: Annexin A1 plays an important role in glioblastoma's proliferation,migration ability,invasiveness,apoptosis and radiosensitivity and takes a part in the activation of NF?b signaling pathway.Annexin A1 may serve as a potential therapeutic target for glioblastoma's treatment in the future.
