Objectives Objective to detect the expression of PD-1/PD-L1 and mRNA in paraffin specimens of non-specific invasive breast cancer,analyze the relationship between PD-1/PD-L1 expression and clinicopathological characteristics of breast cancer,and explore the clinical significance of RNAscope technology in detecting breast cancer PD-1/PDL1 mRNA.Methods A total of 60 cases of non special invasive breast cancer and 30 benign lesions of the mammary tissues were collected.The clinical data of the selected cases were complete.The expression of PD-1/ PD-L1 protein and PD-1 / PD-L1 mRNA in paraffin specimens were detected by immunohistochemistry and RNAscope technique respectively.Results In 60 cases of non special invasive breast cancer cases,the expression of PD-L1 / PD-1protein in the positive rate was 55%(33/60)and 28.3%(17/60),while the positive rate of PD-1/PD-L1 protein in benign breast tissues was 0%(0/30),the difference was statistically significant(P < 0.05).In 60 cases of non special invasive breast cancer,PD-1 protein was not related to age,family history,tumor size,HER2 gene status,TNM stage and molecular typing(P > 0.05).PD-1 protein is associated with histological grading and lymph node metastasis in patients.The positive rate of histological grade III was higher than that(90%,9/10)histologicalgrade II(48%,24/50),statistical analysis the difference was significant(P=0.037);in lymph node metastasis group,the positive rate of metastasis(75%,24/32)than those without metastasis positive expression rate(32.1%,9/28)statistical analysis,the difference was significant(P=0.001).In 60 cases of non special invasive breast cancer,PD-L1 protein was not related to age,family history,tumor size,TNM stage and molecular typing.The expression of HER2 was correlated with HER2 gene status,histological grade and lymph node metastasis in patients with PD-L1.The positive expression with HER2 gene amplification rate(50%,9/18)is higher than the positive rate of HER2 gene amplification(19%,8/42),with a significant difference between the two groups(P=0.015);60% grade school organizations positive expression rate(6/10)expression is higher than that of histological grade II was 22%(11/50)statistical analysis,there was significant difference between the two groups(P=0.040);lymph node metastasis rate was 40.6%(13/32)higher than that without lymph node metastasis was 14.3%(4/28),statistical analysis,a significant difference between the two groups(P=0.049).In 60 cases of non special invasive breast cancer cases,the expression of PD-L1 /PD-1mRNA,the positive rate was 56.7%(34/60)and 30%(18/60),and PD-1/PDL1 mRNA in breast benign lesions were negative,the positive rate was 0%(0/30),there was significant difference in statistical significance(P < 0.05).In 60 cases of non special invasive breast cancer,PD-1mRNA was not related to age,family history,tumor size,HER2 gene status,TNM stage and molecular typing(P > 0.05).It was associated with histological grading and lymph node metastasis of the patients.The positive rate of III(100%,10/10)organization,positive rate is higher than the histological grade II(48%,24/50),statistical analysis the difference was significant(P=0.003);the positive rate of lymph node metastasis(78.1%,25/32)than those without metastasis(32.1%,9/28),there was significant statistical analysis the difference between the two(P=0.000).In 60 cases of non special invasive breast cancer,PD-LmRNA was not related to age,family history,tumor size,TNM stage and molecular typing.The correlation between HER2 and HER2 gene status,histological grade and lymph node metastasis in patients with P was significant.The expression of HER2 associated with gene amplification rate(55.6%,10/18)was higher than that of HER2 gene amplification rate(19%,8/42),with a significant difference between the two groups(P=0.005);the positive rate of histological grade III(70%,7/10)was higher than that of histological grade II was 22%(11/50).Statistical analysis the difference was significant(P=0.008);the positive expression with lymph node metastasis rate(43.8%,14/32)higher than the positive rate without metastasis(14.3%,4/28),statistical analysis of a significant difference between the two groups(P=0.028).60 cases of non immune group special type of invasive breast cancer and RNAscope detection of PD-1 were positive in 31 cases,24 cases were negative,immunohistochemical detection for RNAscope detection for positive and negative in 2 cases,immunohistochemical detection was negative and RNAscope technology for detection.There are 3 positive cases,two had no significant difference(P>0.05)method for detection results,the consistency of two kinds of detection methods is very high(Kappa = 0.831),60 cases were non special type of invasive breast cancer and RNAscope detection of PD-L1 were positive for 16 cases were negative.In 41 cases,immunohistochemical detection for RNAscope detection for positive and negative in 1 cases,immunohistochemical detection was negative but RNAscope positive in 2 cases,two had no significant difference(P>0.05)method for detecting results,two kinds of detection is very consistent High(Kappa value is 0.879).Conclusions The expression of pd-1 / pd-l1 protein and the positive rate of pd-1 / pdl1 mrna expression in non-special invasive breast cancer patients were significantly higher than that of benign lesions,and the expression of pd-1 / pd-l1 may be involved in the occurrence and development of breast cancer.A special type of invasive breast cancer in patients with PD-protein,PD-1 mrna expression is related to histological grade and lymph node metastasis in patients with PD-L1 protein,PD-L1 mRNA expression of HER2 gene amplification with patients,histological grade and lymph node metastasis,prompt PD-1 / PD-L1 prognosis of breast cancer and the biological activities has a certain impact.RNAscope technology and immunohistochemical detection of non-specific invasive breast cancer pd-1 / pd-l1 expression have good consistency,suggesting that RNAscope technology can be used for screening of breast cancer pd-1 / pd-l1 mRNA.
