The Study Of Mongonlian Medicine Zhongluna Decoction On CIA Rats Of OPG And RANKL

Objective To clear the target which is rheumatoid arthritis medical intervention,to provide new strategies for targeting therapy of rheumatoid arthritis and to provide scientific evidence for Mongolian medicine to standardized treatment of rheumatoid arthritis at the same time through the impact of Mongolian medicine Zhonglun-A potions on OPG/RANKL that exists in collagen-induced arthritis(CIA)rat serum or arthritis.Method The experiment arthritis rat model was established by immunized male rats bovine II collagen(B II C),and incomplete Freund's adjuvant(CFA),then divided the mode into normal group,model group,Zhonglun-A potions group(divided into high,medium and low dosage)and Tripterygium Glycosides group which is positive control drug.Observe the general activity,arthritis index and the change of foot thickness on a regular basis during the experiment.Content of anti-CII antibodies,OPG and RANKL in rat serum assayed by ELISA.The knee synovial tissues of the rats were taken and synoviocytes were separated and cultured,synovial cell proliferation was evaluated by the MTT assay.Observe the change of synovial and cartilage tissue 's pathology morphology through the pathological section of ankle joint which are stained with HE,and the OPG and RANKL's expression were detected by immunohistochemistry S-P method.Result CIA rats have clinical situation with obviously persistent swelling oflocal joints,there is higher expressions of anti-CII antibodies,that were significant differences compared with normal group(P<0.05).The drugs can increase weight,relieve rat paw edema,arthritis index and the pathological morphology changes.The OPG(in rats serum and ankle joint)is higher expression and RANKL(in rats serum and ankle joint)is lower expression in normal group,but OPG is lower expression and RANKL is higher expression in model group,there was significant difference between the two groups(P<0.05),the drug therapy groups were all can lower the value of RANKL and higher OPG,thus increasing the OPG/RANKL ratio,there was significant difference with the model group(P < 0.05),among them the Tripterygium Glycosides and the Mongolian medicine are the best,there was no significant difference between them(P>0.05),the medium and low dosage of Mongolian medicine are poorer thanthe Tripterygium Glycosides(P < 0.05).Conclusion Zhonglun-A potions has obvious improvement of CIA rat in general condition,and it can obviously improve rat's diet and gain them weight,to a certain extent to relieve arthritis symptoms and reduce the lesion rats foot thickness,a measurable increment higher OPG and lower RANKL both in serum and arthrosis,thus increasing the OPG/RANKL ratio.Zhonglun-A potions mainly about to hold-up the expression of RANKL to higher the expression of OPG,so that to increasing the OPG/RANKL ratio,through competitive binding that OPG can interdict combine of RANK and RANKL,to implement the ease or delay RA's progression of disease.This may be one of Mongolian medicine Zhonglun-A potions' mechanism for the treatment of rheumatoid arthritis.
