Cancer is one of the main diseases of a serious threat to human health,at present the incidence of cancer is increasing by an average 3%-5%.Lung cancer is the fastest growing malignant tumor of morbidity and mortality in the worldwide,it has become the first disease of the malignant tumors in many developed countries and some developing countries.The three conventional treatment methods are surgery,chemotherapy and radiotherapy;besides,there are some new treatments methods,such as immune therapy,gene therapy and so on.However,surgical resection could not clean up all tumor cells,it also can't remove the cause of cancer,the same as it has postoperative recurrence and metastasis problems,while radiation therapy,chemotherapy because of the serious adverse reaction restrain their application.Visibly,these methods of treatment of tumor have limitations and incompleteness,they will not only cause serious side effects,but cannot fundamentally solve the tumor recurrence,metastasis and chemical resistance.Photodynamic therapy can induce apoptosis and necrosis by damage cell structure or affect cell function to produce therapeutic effect;at the same time it has highly effective,minimally invasive and low cost characteristics;So the new type photodynamic therapy of small trauma is aroused people's wide attention and research,and become a new direction in the field of tumor treatment.In this article,we elaborate the experimental research that photodynamic therapy of fiber import within the tumor by percutaneous puncture treat lung cancer,the main research content is as follows:1.The structure of tumor model: nude mice inoculate A549 lung cancer cell on the back shoulder blades to structure tumor model,inoculating tumor cells after one week,subcutaneous transplantation tumor appeared visibly,the shape is circular or elliptic nodules,these information mean success of nude mouse lung cancer model building;according to different photosensitizer dosages(5 mg/kg,15 mg/kg and 30 mg/kg),treatment groups are divided into three group to treat lung cancer with photodynamic therapy of fiber import within the tumor by percutaneous puncture;after completion of treatment,analyses the changes of tumors volume and inhibitory rate;experimental results show that the photodynamic therapy of fiber import within the tumor by percutaneous puncture for volume growth of tumors has obviously inhibitory effect,which prompted photodynamic therapy for the treatment of lung cancer have a positive impact.2.The action mechanism of photodynamic therapy for the treatment of tumor is complex,after the experiment,we got tumors organization,through the observation of the pathological,VEGF and b-FGF expression level of detection in tumors organization,analysis of the experimental results;the results showed that the light dose and illumination time under the condition of the same,with the increase of concentration of photosensitizer,the degree of cell death in tumor tissue is greater,the expression level of VEGF and b-FGF is lower,these show photodynamic therapy can kill tumor cells,inhibiting angiogenesis factor expression,to achieve the effect of treatment.
