As a serious disease,cancer has been harmful to human beings for a long time.So far,most cancers are still unsolved problems for human beings.Leukemia,as a common malignant tumor,has a high mortality rate,a wide coverage of the age of onset,and its incidence is increasing year by year.Studies have shown that the generation and progression of leukemia are related to the abnormal expression of many genes.MicroRNA(miRNA)plays an important role in the regulation and expression of gene.People have been trying to find molecular biomarkers in the stage of leukemia disease for a long time,hoping to overcome the problem.However,because of the heterogeneity and complexity of leukemia,its development is not only related to the individual biomarker,but also related to the interaction between multiple biomolecular modules.Therefore,from the gene network level,the search for multiple biomolecular markers is more robust than a single marker,and has more general applicability for different individuals.The main subjects of this study are Acute Myelocytic Leukemia(AML)and Chronic Myelocytic Leukemia(CML).First,we collected data from miRNA expression microarray of AML and CML from gene expression database GEO(Gene Expression Omnibus).Using the LIMMA algorithm to calculate the difference miRNA of two types of leukemia,we use the MINET algorithm to calculate the correlation coefficient between different miRNA,and then use the difference miRNA as the breakthrough point to filter out the strong correlation interaction pairs with different miRNA.On the basis of this,construct the miRNA co-expression network of two types of leukemia by Cytoscape software,through the analysis of the topology of the abnormal co-expression network,find the miRNA that plays a central role in it.Using two kinds of software MiRanda and Targetscan to predict the target gene of the core miRNA,and then combine the miRNA target gene of the MiRTarBase database to obtain the target gene of the core miRNA.Finally,we combine KEGG and GO gene annotation results with specific literature to compare and analyze two kinds of leukemia.These computer methods are used to analyze biological data and to propose molecular and regulatory relationships associated with the development of leukemia,providing a theoretical basis for the diagnosis and treatment of two types of leukemia and a new biological target.At last,there were 5 different expressions of miRNA in two types of leukemia,32 abnormal expression of 32 genes regulated by 14 miRNA in AML,and 29 abnormal expression of 11 miRNA regulated in CML.
